摘  要:【目的】从川南丰富的高粱种质资源中筛选酿酒专用高粱材料并进行差异分析,旨在解决主栽品种退化及品种同质化问题。【方法】利用超高效液相色谱—串联质谱技术从22份高粱种质资源材料中筛选出川南高粱地方品种HS08,分析其与国窖红1号、红缨子、朗糯红19号3种代表性酿酒专用常规品种的代谢物组成差异,并结合理化组分以及产量性状进行分析,用以辅助新品种选育。【结果】高粱地方品种HS08的单穗籽粒重(87.73 g)及千粒重(21.13 g)显著高于3个常规品种平均值(单穗重68.50 g、千粒重18.79 g)。HS08的淀粉、蛋白质、脂肪、单宁、直链淀粉等含量适中,与3种常规品种的理化成分无显著差异,符合酿酒高粱标准。4种高粱检测出18类共2421个代谢物。HS08相较于国窖红1号、红缨子及朗糯红19号分别筛选出81、62、74个差异代谢物,主要包括黄酮类、萜类、香豆素类、酚酸类等次生代谢物,以及脂类、氨基酸类及其衍生物等初生代谢物。差异代谢物中,黄酮类占比最高且多为上调。对代谢物进行富集分析显示,显著富集且上调的共有代谢通路有类黄酮生物合成、黄酮和黄酮醇生物合成。【结论】地方品种HS08富含黄酮类等抗氧化活性物质,具有更高营养价值和生物抗性,理化指标适中,可用以培育优质高粱。利用理化分析和非靶向代谢组学相结合的方法可为酿酒高粱新品种筛选和选育提供理论依据。【Objective】The paper aimed to solve the problems of degradation and homogenization of the main cultivated varieties,special sorghum materials for brewing were selected from the abundant sorghum germplasm resources in southern Sichuan,and differential analysis was conducted.【Method】The sorghum landrace HS08 in southern Sichuan was filtered from 22 sorghum germplasm resources using ultra-high performance liquid chromatography tandem mass spectrometry technology.The differences in metabolite composition as well as the physicochemical components and yield traits of the HS08 and the three conventional varieties for brewing including Guojiaohong No.1,Hongyingzi,and Langnuohong No.19 were analyzed for use in the screening of sorghum materials for brewing and the selection of new varieties which aimed at solving the issue of degradation of main crop varieties.【Result】The grain weight(87.73 g) and thousand grain weight(21.13 g) of HS08 were significantly higher than the average of the three conventional varieties(68.50 g and 18.79 g).There was no significant difference in the physicochemical compositions of the four sorghum varieties,and the starch,protein,fat,tannin,and amylose contents of HS08 met the standard requirements for brewing sorghum.A total of 2421 metabolites belonging to 18 categories were detected in the four sorghums.There were significant differences in the metabolome of HS08 compared to Guojiaohong No.1,Hongyingzi,and Langnuohong No.19,and 81,62 and 74 differential metabolites were screened out respectively,which mainly included secondary metabolites such as flavonoids,terpenoids,coumarins and derivatives,phenolic acids and derivatives,steroids and derivatives,as well as primary metabolites such as lipids,amino acids and derivatives,etc.Among the three groups of differential metabolites,flavonoids were the most highly represented and mostly up-regulated.The shared metabolic pathways that were significantly enriched and up-regulated were flavonoid biosynthesis,flavonoid and flavonol biosynthes
