摘 要:【目的】探究发情与未发情西门塔尔母牛血液代谢物和子宫颈菌群结构的变化、血液代谢物和菌群变化以及生殖激素和菌群变化之间的潜在关联。【方法】通过酶联免疫吸附实验分别检测6头发情和6头未发情西门塔尔肉用母牛血液生化指标、生殖激素水平,同时基于16 SrRNA基因高通量测序技术和UPLC-MS/MS代谢组学技术分析发情和未发情母牛子宫颈菌群结构变化。【结果】(1)与未发情组相比,发情组母牛血液中总蛋白、球蛋白和葡萄糖含量极显著升高(P<0.01,下同),白蛋白、胆固醇、雌二醇和胰岛素样生长因子显著升高(P<0.05,下同),孕酮含量显著降低。(2)与未发情组相比,发情母牛血液中181种代谢物发生显著变化(VIP≥1,FC≥1.5或FC≤1/1.5),其中84种上调,97种下调。差异代谢物显著富集于精氨酸和脯氨酸代谢、氨基酸的生物合成、蛋白质的消化和吸收等11条代谢通路。(3)与未发情组相比,在门水平上,子宫颈厚壁菌门和拟杆菌门丰度均极显著上升;在属水平的优势菌群主要为肠道细菌,其次是拟杆菌属。(4)拟杆菌属的丰度与肌酸呈极显著负相关,与L-丙氨酸、吲哚-3-乙酸、γ-氨基丁酸呈显著负相关;肠单胞菌属的丰度与γ-氨基丁酸、N-乙酰羟色胺呈极显著负相关,与吲哚-3-乙酸呈显著负相关。(5)拟杆菌属的丰度与胰岛素样生长因子(IGF-1)呈显著正相关,unclassified_Firmicutes的丰度与IGF-1呈显著负相关。【结论】母牛发情后血液生化指标、子宫颈细菌结构和血液代谢物发生显著变化,且血液代谢物变化和生殖激素变化与菌群结构密切相关。【Objective】The present paper aimed to investigate the changes in blood metabolites and cervical microbiota structure,as well as potential associations between changes in blood metabolites and microbiota,reproductive hormones,and microbiota in estrus and non-estrus Simmental cows.【Method】Enzyme linked immunosorbent assay was used to detect blood biochemical indicators and reproductive hormone levels in 6 estrus and 6 non-estrus Simmental meat cows,respectively.At the same time,high-throughput sequencing technology of 16 SrRNA gene and UPLC-MS/MS metabolomics technology were used to analyze the structural changes of cervical microbiota in estrus and non-estrus cows.【Result】(i) Compared with the non-estrus group,the estrus group showed an extremely significant increase in total protein,globulin,and glucose content in the blood of cows(P<0.01,the same as below),as well as a significant increase in albumin,cholesterol,estradiol,and insulin-like growth factor(P<0.05,the same as below),and a significant decrease in progesterone content.(ii) Compared with the non-estrus group,there were significant changes in 181 metabolites in the blood of estrus cows(VIP ≥ 1,FC ≥ 1.5 or FC ≤ 1/1.5),with 84 up-regulated and 97 down regulated.Significant enrichment of differential metabolites included 11 metabolic pathways,including arginine and proline metabolism,amino acid biosynthesis,protein digestion and absorption.(iii)Compared with the non-estrus group,the abundance of Firmicutes and Bacteroidetes in the cervix significantly increased at the phylum level;The dominant microbial community at the genus level was mainly composed of intestinal bacteria,followed by Bacteroides.(iv)The abundance of Bacteroides was extremely significantly negatively correlated with creatine,and significantly negatively correlated with L-alanine,indole-3-acetic acid,and gamma aminobutyric acid;The abundance of Enteromonas was extremely significantly negatively correlated with gamma aminobutyric acid and N-acetylserotonin,and significantly
