摘 要:植物在应对非生物胁迫时,脱落酸(ABA)发挥着关键的调节作用。生物体合成ABA的过程中9-顺式-环氧类胡萝卜素加双氧酶蛋白(9-cis-epoxycarotenoids dioxygenase,NCED)具有重要调控作用。本研究通过生物信息学方法开展苹果NCED基因家族成员鉴定,进而通过表达图谱分析了在干旱和盐胁迫条件下苹果MdNCEDs基因的表达情况。结果表明,本研究鉴定得到MdNCED基因家族成员18个,染色体定位分析显示18个MdNCEDs分布在10条染色体上;理化性质分析显示MdNCEDs分子量为46.47-76.36 kD,等电点介于5.2-7.63之间。苹果MdNCED启动子序列中检测到多种与逆境激素、响应非生物胁迫反应功能元件;基因结构分析显示,MdNCED基因结构有较高一致性;共线性分析结果显示,部分基因之间发生了复制事件;苹果MdNCEDs基因响应干旱、高盐胁迫的表达数据显示,与CK(0 h)相比,在高盐和干旱两种非生物胁迫处理下3个基因(MdMYB3、MdMYB11、MdMYB18)上调表达,4个基因(MdMYB1、MdMYB6、MdMYB13、MdMYB17)下调表达,2个基因(MdMYB4、MdMYB14)表达无明显规律,4个基因(MdMYB5,MdMYB8,MdMYB9,MdMYB16)无显著性差异。本文为后续深入解析苹果MdNCEDs基因的功能提供了基础理论依据。Abscisic acid(ABA)plays a crucial role when plants are coping with abiotic stresses.NCED exerts important regulatory effects in the synthesis of ABA in organisms.This research analyzes the members of apple NCED gene family through bioinformatics methods,then detects the expression of apple MdNCEDs under different drought and salt stress conditions using expression profiles.The results show that 18 MdNCED gene family members are identified in this study.Chromosome localization analysis indicates that these members are distributed on 10 chromosomes.The molecular weight of MdNCEDs is 46.47-76.36 kD,and the isoelectric point(pI)is between 5.2-7.63.A variety of functional elements relates to stress hormones and responses to abiotic stress are detected in apple MdNCED promoter sequence.Gene structure analysis reveals that the structure of MdNCED gene is highly consistent.The results of collinearity analysis demonstrat that duplication events occur between some genes.The expression data shows that compared with CK(0 h),three genes(MdMYB3,MdMYB11,MdMMYB18)are up-regulated,and four genes(MdMYB1、MdMYB6、MdMYB13、MdMYB17)are down-regulated under high-salt and drought stress conditions.There is no obvious regularity in the expression of MdMYB4 and MdMYB14 or significant differences in the four genes(MdMYB5,MdMYB8,MdMYB9,MdMYB16).This paper provides a basic theoretical basis for further analysis of the function of MdNCED genes in apple.
