摘 要:按照处方量制备宣肺败毒方,武火煮沸后,文火煎煮60 min,纱布过滤,文火浓缩煎煮至400 m L,滤纸过滤,再用0.45μm水系滤膜过滤,在Agela Promosil C_(18)色谱柱上进行分离,在柱温30℃条件下以不同体积比的0.1%(体积分数)磷酸溶液-甲醇混合溶液为流动相进行梯度洗脱,在波长220 nm处检测,记录色谱图。根据中药色谱指纹图谱相似度评价系统(2004 A版)建立了宣肺败毒方的高效液相色谱(HPLC)指纹图谱。结果显示,11批宣肺败毒方指纹图谱中有24个共有峰,与大黄素对照品比对,确认24号峰对应大黄素,11批宣肺败毒方指纹图谱与对照指纹图谱的相似度为0.988~1.000。According to the prescribed amount,the Xuanfei Baidu formula was prepared.After boiling with flamming fire,the mixture was simmered for 60 min,passed through gauze,and concentrated its volume up to 400 mL by soft fire.The mixture was passed through filter paper and 0.45μm water filter membrane.The test solution of Xuanfei Baidu formula was separated on Agela Promosil C_(18) chromatographic column,using the mixed solution composed of 0.1%(volume fraction)phosphoric acid solution and methanol at different volume ratios as mobile phase for gradient elution at 30℃of column temperature.The components of the test solution were detected at wavelength of 220 nm,and the chromatogram was recorded.High performance liquid chromatography(HPLC)fingerprint of Xuanfei Baidu formula was established according to the Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(2004 A Edition).As shown by the results,there were 24 common peaks in the fingerprints of 11 batches of Xuanfei Baidu formula.Compared with the emodin reference substance,the compound of No.24 peak was confirmed to be emodin,and the similarity was in the range of 0.988-1.000 between fingerprints of 11 batches of Xuanfei Baidu formula and reference fingerprint.
