扶正辟邪方对新冠病毒S蛋白联合LPS致小鼠系统性炎症反应影响的研究

摘　　要：目的评价扶正辟邪方调节免疫功能的作用,为其临床应用提供科学的参考依据。方法利用多次注射新冠病毒S蛋白后再联合脂多糖(LPS)诱导小鼠产生多系统炎症,腹腔注射环磷酰胺建立免疫功能低下小鼠模型,考察扶正辟邪方对机体免疫功能的调节作用。结果扶正辟邪方显著升高白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、单核细胞趋化蛋白-1(MCP-1)、巨噬细胞炎性蛋白-1α(MIP-1α)和调节激活正常T细胞表达分泌因子(RANTES)水平,减轻系统性炎症反应小鼠肺间质炎细胞浸润以及肺间质增宽程度,改善小鼠脾脏白髓增生、副皮质区萎缩、红髓萎缩和星空细胞增生,抑制小鼠体温降低;扶正辟邪方可以显著增加小鼠巨噬细胞的吞噬能力。结论扶正辟邪方可显著改善S蛋白及LPS诱导系统性炎症,可有效预防新冠病毒感染,其作用机制可能与其具有调节细胞因子风暴、调节免疫功能等作用有关。Objective To evaluate the effect of Fuzheng Pixie Recipe(扶正辟邪方)on regulating immune function and provide scientific reference for its clinical application.Methods Using multiple injections of S protein of novel coronavirus combined with lipopolysaccharide(LPS)to induce multisystem inflammation in mice,and intraperitoneal injection of cyclophosphamide to establish an immunosuppressive mouse model to investigate the regulatory effect of Fuzheng Bixie Recipe on the immune function of the body.Results Fuzheng Pixie Formula significantly increased the levels of IL-6,IL-10,MCP-1,MIP-1αand RANTES,it reduces the infiltration of interstitial inflammatory cells and non interstitial widening in the lungs of mice with systemic inflammatory response,improves splenic white pulp hyperplasia,paracortical atrophy,red pulp atrophy,and starry cell proliferation,and inhibits the decrease of mouse body temperature.Fuzheng Pixie Formula can significantly increase the phagocytic capacity of mouse macrophages.Conclusion Fuzheng Pixie Recipe can significantly improve systemic inflammation induced by S protein and LPS,and can effectively prevent novel coronavirus infection.Its mechanism may be related to its role in regulating cytokine storm and immune function.
