摘 要:目的探讨大黄■虫丸干预肺纤维化(PF)的相关机制。方法选取8周龄SPF级雄性SD大鼠20只,体重(300±10)g,按照计算器法分为空白组、大黄■虫丸组,每组10只。大黄■虫丸组灌胃给药0.45 g/(kg·d),1次/d;空白组每日给予等体积的去离子水,连续给药7 d。使用UPLC-Q-TOF-MS技术对大黄■虫丸入血成分做定性分析,并结合网络药理学方法,分析大黄■虫丸干预PF的核心成分、靶点和通路,绘制网络图。结果筛选出大黄■虫丸入血活性成分27种;核心成分包括喹啉类生物碱、二萜、黄酮、蒽醌及其他类,得到16个核心靶点,包括TNF、MAPK、mTOR、PI3Ks、MMPs等;治疗PF的核心通路涉及病毒、肿瘤、能量代谢、血管病变等。结论本研究初步预测了大黄■虫丸干预PF的主要入血成分及相关靶点和通路,其机制可能与细胞增殖、分化、迁移、氧化应激、感染等有关。Objective To explore the mechanism of Dahuang Zhechong Pills in intervening pulmonary fibrosis(PF).Methods A total of 20 male SD rats of 8-week-old SPF grade,weighting(300±10)g were selected,and they were divided into blank group and Dahuang Zhechong Pills group according to the calculator method,with 10 rats in each group.Dahuang Zhechong Pills group was administered by intragastric administration 0.45 g/(kg·d),one time a day;and blank group was given an equal volume of deionized water daily,the drug was administered continuously for 7 days.UPLC-Q-TOF-MS technique was used to qualitatively analyze the blood components of Rhubarb Zhechong Pills,and combined with network pharmacology methods,the core components,targets,and pathways of Dahuang Zhechong Pills to interfere with PF were analyzed,and the network diagram was drew.Results A total of 27 active ingredients of Dahuang Zhechong Pills were screened;the core components included quinoline alkaloids,diterpenoids,flavonoids,anthraquinones and others,and 16 core targets were obtained,including TNF,MAPK,mTOR,PI3Ks,MMPs,etc.The core pathway of PF treatment involves virus,tumor,energy metabolism,vascular disease and so on.Conclusion This study preliminarily predicted the main blood-entry components and related targets and pathways of Dahuang Zhechong Pills for intervening in PF,and its mechanism of action may be related to cell proliferation,differentiation,migration,oxidative stress,and infection.
