摘  要:少于6 h的短程运输是牛卵巢采集后体外胚胎生产(IVP)的核心环节,温度选择将直接影响卵母细胞发育。试验在4~6 h短程运输条件下,对比15℃与37℃温度条件对卵母细胞成熟及胚胎发育的影响。结果表明:37℃组卵母细胞成熟率与囊胚形成率显著高于15℃组(P<0.05),复温后37℃组HSP70表达量更高,且复温4 h后37℃组卵母细胞的丙二醛(MDA)浓度显著低于15℃组(P<0.05)。说明短程运输中37℃与15℃相比更有利于维持生理代谢与结构完整性,能够显著提升卵母细胞质量,推荐作为标准化操作温度。Short-term transportation(<6 h)of bovine ovaries is a critical step in post-collection in vitro embryo production(IVP),where temperature setting directly influences oocyte developmental competence.This study simulated 4-6 h transportation conditions to compare the effects of 15℃and 37℃on oocyte maturation and embryonic development.Results demonstrated that the significantly higher oocyte maturation rates[(83.2±3.1)%vs.(64.7±4.5)%,P<0.05]and blastocyst formation rates[(42.6±2.8)%vs.(28.3±3.4)%,P<0.05]were observed in the 37℃transportation group,compared to the 15℃group.Post-rewarming analysis revealed elevated HSP70 expression in the 37℃group,along with lower malondialdehyde(MDA)concentrations in oocytes after 4 h of temperature recovery[(1.8±0.3)nmol/mg vs.(3.2±0.4)nmol/mg,P<0.05].These findings indicated that a better physiological metabolism and structural integrity during short-term transportation for collected oocytes could be achieved with the transportation temperature of 37℃,compared to 15℃.Therefore,37℃is recommended as the standardized operational temperature for short-term transportation of oocytes in IVP.
