摘 要:为探索昆虫资源高值化利用,本试验以蝇蛹为原料提取蛋白质和制备甲壳素。采用碱提法、Box-Behnken设计、响应面优化脱蛋白工艺酸沉提取蛋白质,酸浸法脱盐和氧化法脱色制备甲壳素,同时红外光谱分析进行表征。结果表明:碱提法提取蝇蛹蛋白质最佳条件为NaOH浓度10.5%,反应温度100℃,反应时间3.2 h,料液比1:20,蛋白质提取率为(50.26±0.20)%。酸浸法脱盐浓度为2 mol/L HCl;氧化法脱色条件为1%NaClO。红外光谱分析发现,制备甲壳素显示典型特征吸收峰,甲壳素提取率为18.89%。本研究可为蝇蛹或其他昆虫资源提取蛋白质和甲壳素提供参考。In order to explore the high-value utilization of insects,fly pupae was used as raw material for extracting protein and preparing chitin in this experiment.The deproteinization process was optimized using Box-Behnken design of response surface method for extracting protein by alkaline extraction and acid precipitation,while the desalination and decolorization process was performed by acid leaching and oxidation method for preparing chitin,respectively,and the characterization was also conducted by infrared spectrum analysis.The results showed that the optimal conditions by alkaline extraction for extracting protein were as follows:NaOH concentration of 10.5%,reaction temperature of 100℃,reaction time of 3.2 h,solid-liquid ratio of 1:20,and the extracted yield of protein was(50.26±0.20)%.The optimal desalination concentration by acid leaching method was 2 mol/L HCl.The decolorization condition by oxidation method was 1%NaClO.Infrared spectrum analysis of prepared chitin showed a typical characteristic absorption,and the final yield of chitin was 18.89%.This study could provide a reference for extracting protein and chitin from fly pupae or other insects resource.
