摘 要:为开发新型生物碱添加剂,本试验以博落回果荚为原料,通过响应面法优化乙醇提取博落回总生物碱的工艺条件,并结合大孔树脂法进行分离纯化。结果表明:最佳提取工艺为:乙醇浓度70%,料液比1:16(g/mL),提取时间2 h,提取温度70℃。经3次平行试验得到黔产博落回生物碱平均提取得率为11.98%,其理论预测值为12.02%,相对误差0.33%,表明该模型可靠。优选出HPD100为洗脱树脂,并采用梯度洗脱得到四种纯化的成分,通过HPLC对纯化后的生物碱纯度进行鉴定,结果显示纯化后的总生物碱纯度超过70%,其中盐酸血根碱为68.2%,盐酸白屈菜红碱为8.5%。研究表明,乙醇法能有效提取博落回中的生物碱,大孔树脂分离使得生物碱纯度提高,丰富了博落回生物碱利用的途径。The experiment aimed to develop new types of alkaloid additives.Macleaya cordata(Willd.)R.Br.fruit pods were used as raw materials,the process conditions for ethanol extraction of total alkaloids from Boluohui were optimized by response surface methodology,and combined with macroporous resin method for separation and purification.The results showed that the optimal extraction process was as follows:ethanol concentration of 70%,solid-liquid ratio of 1:16(g/mL),extraction time of 2 hours,and extraction temperature of 70℃.After three parallel experiments,it was found that the average extraction rate of alkaloids from Macleaya cordata(Willd.)R.Br.in Guizhou was 11.98%,with a theoretical prediction value of 12.02%and a relative error of 0.33%,indicating the reliability of the model.HPD100 was selected as the elution resin,and four purified components were obtained by gradient elution.The purity of the purified alkaloids was identified by HPLC,and the results showed that the total purity of the purified alkaloids exceeded 70%,including 68.2%of sanguine hydrochloride and 8.5%of berberine hydrochloride.Research had shown that the ethanol method could effectively extract alkaloids from Macleaya cordata,and macroporous resin separation improves the purity of alkaloids,enriching the pathways for the utilization of Macleaya cordata(Willd.)R.Br.alkaloids.
