摘 要:通过人工气候箱模拟低温高湿环境建立寒湿困脾证泄泻小鼠模型,并依据“以方测证”法应用藿香正气散对模型进行验证。实验将20只小鼠随机分为正常组(n=10)和模型组(n=10),模型组小鼠置于温度(4±0.5)℃、湿度(90±2)%的人工气候箱中,4 h/d,连续7 d,通过观察小鼠寒湿困脾证泄泻主次症来评估模型,并检测血脂含量、氨基酸代谢相关酶活性;然后,另将30只小鼠分为正常组(n=10)和模型组(n=20),造模成功后将模型组进一步分为自然恢复组和藿香正气散组(n=10)。藿香正气散组灌胃藿香正气散水煎液,其他组灌胃等量无菌水,观察疗效。造模结果显示,与正常组比,模型组小鼠稀便率和粪便含水量升高(P<0.01),肛温和腹温降低(P<0.01),其症状符合寒湿困脾证泄泻的主次症辨证诊断标准;模型组小鼠出现血清血脂含量及氨基酸代谢相关酶活性的变化。治疗结果显示,藿香正气散组小鼠稀便率降低(P<0.05),腹温升高(P<0.05);该组小鼠血清血脂含量及氨基酸代谢相关酶活性接近正常组水平。基于模型评估结果及藿香正气散验证结果,判定模拟低温高湿环境成功建立了寒湿困脾证泄泻小鼠模型。Objective:The mouse model of diarrhea with the cold-dampness impairing spleen syndrome was established through simulating the low-temperature and high-humidity environment.The model was validated using Huoxiang Zhengqi San based on the“using formulas to validate syndromes”approach.Methods:Twenty Kunming mice were randomly assigned to normal(n=10)and model groups(n=10).Mice in the model group were exposed to an artificial climatic chamber set at(4±0.5)°C and(90±2)%humidity for four hours per day over seven consecutive days.Symptoms of diarrhea with cold-dampness impairing spleen syndrome were evaluated,and blood lipid levels along with the activity of enzymes involved in amino acid metabolism were measured.Thereafter 30 mice were divided into two groups:normal group(n=10)and model group(n=20).Through modeling,the model group was further divided into a natural recovery group and a Huoxiang Zhengqi San treatment group(n=10).The treatment group was administered Huoxiang Zhengqi San decoction via gavage,while the other groups received an equal volume of sterile water.Therapeutic efficacy was observed.Results:The modeling results showed that,compared with the normal group,the model group had increased rates of loose stools and fecal water content(P<0.01),and decreased rectal and abdominal temperatures(P<0.01),meeting the diagnostic criteria for the primary and secondary symptoms of diarrhea with cold-dampness impairing spleen syndrome.The model group also displayed changes in serum lipid levels and the activity of enzymes involved in amino acid metabolism.The treatment results showed that the Huoxiang Zhengqi San group had a reduced rate of loose stools(P<0.05)and increased abdominal temperature(P<0.05).Serum lipid levels and the activity of enzymes involved in amino acid metabolism in the treatment group were comparable to those in the normal group.Conclusion:Based on the model evaluation results and the verification findings of Huoxiang Zhengqi San,the mouse model of diarrhea was successfully established withi
