天麻植物纳米囊泡的制备及其体外经皮渗透

摘  要:通过制备天麻植物纳米囊泡(Gastrodia Elata Plant-drived Nanovesicles,GEPDNVs),得到平均粒径为89nm、具有明显脂质双分子层膜结构的圆形囊泡,并且通过高效液相色谱法测定天麻素的质量浓度为0.06%。在B16黑素瘤细胞实验中,在≤10%质量分数情况下,细胞存活率均大于80%,并且对酪氨酸酶的抑制率呈剂量依从赖性。根据qPCR基因表达测试结果,1%和10%浓度的GEPDNVs对三种调控黑色素生成的关键基因均有显著抑制作用,并且也呈现剂量依赖性。在体外Franz扩散池透皮吸收测试中,设置两个变量:活性物起始浓度和渗透时间,与对照组去离子水相比,GEPDNVs均表现出优异的促渗透性。AGastrodia Elata Plant-drived Nanovesicles(GEPDNVs)with an average diameter of 89nm and an obvious lipid bilayer membrane structure were prepared.The mass concentration of gastrodin was determined by HPLC up to 0.06%.In the B16 melanoma cell experiments,the cell viability was greater than 80%when the mass concentration was less than 10%,and the inhibition rate of tyrosinase was dose-dependent.According to qPCR gene expression test results,1%and 10%GEPDNVs significantly inhibited three essential genes regulating melanin production,and also presented dose-dependent inhibition.In vitro test of transdermal absorption of Franz diffusion cell,GEPDNVs demonstrated excellent permeability compared with deionized water when two variables were set:initial concentration of actives and penetration time.
