双氢青蒿素对患卡氏肺孢子虫肺炎大鼠血清和肺泡巨噬细胞上清液IL-1水平的影响

摘　　要：为研究双氢青蒿素对患卡氏肺孢子虫肺炎 (Pneumocystiscarinii pneumonia,PCP)大鼠血清和肺泡巨噬细胞培养上清液 IL - 1水平的影响 ,以醋酸可的松皮下注射 Wistar大鼠建立卡氏肺孢子虫肺炎动物模型 ,用 6 0 mg/ kg双氢青蒿素治疗实验大鼠 ,杀鼠取肺 ,用胶原酶消化法分离大鼠肺泡巨噬细胞 ,用 L PS刺激培养 72 h,同时设有感染对照组和正常对照 ,用 IL - 1β试剂盒分别检测血清和培养上清液 IL - 1β的水平 ,结果显示感染组和治疗组大鼠 IL - 1β水平显著高于正常对照 ,治疗组大鼠 IL - 1β水平则低于感染组 ,说明卡氏肺孢子虫感染可能引起大鼠肺泡巨噬细胞分泌高水平 IL - 1,经双氢青蒿素治疗 PCP后大鼠肺泡巨噬细胞产生 IL - 1水平降低。To study the effects of treatment with dihydroartemisinin(DHA) on IL 1 in sera and the supernatant of alveolar macrpophages(AMs) in rats infected with Pneumocystis carinii pneumonia(PCP), Wistar rat model of PCP was established by subcutaneous injection witn cortisone acetate, experimental rats were treated by 60mg/kg DHA and killed to obtain lungs, AMs were separated by collagenase digestion and cultured by LPS stimulation in 72 hours,infected and normal rats served as controls, the levels of IL 1β in sera and the AMs' supernatant were detected by Kits of IL 1β respectively. Results showed that the level of IL 1β in the infection and treatment groups was higher than that in the normal control, that in the treatment group was lower than that in the infection group. It could be concluded that AMs of PCP rats secreted a high level of IL 1,but the level of IL 1 was decreased after PCP rats accepted DHA therapy.
