摘 要:为观察蒿甲醚 (Art)对日本血吸虫 (Schistosoma japonicum)超氧化物歧化酶 (SOD)和谷胱甘肽还原酶 (GR)的影响 ,取血吸虫成虫置于含 Art(7.5~ 6 0 μmol/ L)和 /或氯化血红素 (hemin,8~ 5 0 μmol/ L)的培养液内培养 2 4 h后 ,分别按邻苯三酚自氧化和 NADPH氧化方法测定虫体的 SOD和 GR活力 ,结果表明 ,在 hemin(10 μmol/ L)存在下 ,Art(7.5~6 0 μmol/ L)对虫体 GR有明显的抑制作用 ,其效果与使用的 Art浓度有关 ,将 hemin浓度升至 4 0 μmol/ L,Art对虫体SOD亦有明显的抑制作用 ,此外 ,Art明显增强 Fe( )诱导的血吸虫蛋白巯基含量下降 ,提示 Art具有铁依赖的抑制血吸虫 SOD和 GR作用 。In this paper, an attempt was made to study the effect of artemether(Art) on two kinds of important antioxidant enzymes superoxide dismutase (SOD) and glutathione reductase (GR) of Schistosoma japonicum. The paired worms were preincubated in vitro without or with Art (7.5~60μmol/L) and/ or hemin (8~50μmol/L) for 24h, then the GR and SOD activities in the worms were determined by the pyrogallol autoxidation and NADPH oxidation assays, respectively. The results showed that a significant decrease in the GR activities in the worms exposed to Art (7.5~60μmol/L) and hemin (10μmol/L) for 24h. In the presence of a higher hemin concentration (40μmol/L), the SOD activities of the worms were also significantly inhibited by Art in a concentration related pattern. Moreover, Art significantly enhanced Fe(II) induced reduction of the worm protein thiol content. The results suggest that Art' iron dependent inhibition of SOD and GR of schistosomes be related to its protein oxidation activities.
