摘 要:目的 探讨Ⅱ组代谢性谷氨酸受体 (metabotropicglutamatereceptors,mGluRs)激动剂二碳酸环氧丙基甘氨酸 (di carboxycyclopropylglycine,DCG IV)对次声脑损害大鼠皮质神经元损伤的干预作用 .方法 SD大鼠 6 0只 ,随机等分为生理盐水对照组及DCG Ⅳ干预组 ,各组再等分为次声作用 1,7和 14次 3个小组 ,DCG Ⅳ干预组采用立体定向的方法向右侧侧脑室内注射 10 μLDCG Ⅳ ;生理盐水对照组按照相同方法注入等量生理盐水 .实验各组每次在次声压力仓内暴露于 8Hz,130dB次声 2h.采用神经功能损伤程度评分 (neurologicalse verityscore,NSS)和病理组织学技术分别研究各组动物于次声作用后脑皮质功能与结构的变化 .结果 次声作用 7和 14次DCG Ⅳ干预组大鼠NSS分值和皮质损伤神经元的计数均显著低于相同次声作用次数的生理盐水对照组 (P <0 0 1) .并且DCG Ⅳ干预组与相同次声作用次数的生理盐水对照组比较 ,脑皮质组织结构损伤程度减轻 .结论 次声脑损伤大鼠模型中 ,DCG Ⅳ通过活化Ⅱ组mGluRs而有效地干预次声引起的脑组织损伤 .提示DCG Ⅳ能够在次声脑损伤中介导神经保护机制 ,具有一定的应用前景 .AIM To study the effects of Group II mGluRs agonist DCG Ⅳ on the damage of rat's brain cortex induced by infrasound. METHODS 60 SD rats were randomly divided into two groups: shame group and DCG Ⅳ group, and each group was randomly divided into three subgroups:1 time, 7 times and 14 times infrasound exposure groups. Rats of shame were injected in paracele with 10 μL normal saline by stereotactic method and those of DCG Ⅳ group were injected with 10 μL DCG Ⅳ. All the rats were exposed to 8 Hz, 130 dB infrasound for 2 hours each time. The alteration of cortex function and structure was observed by neurological severity score and pathology method. RESULTS Compared with shame groups, both the neurological severity score and the number of injured cortex neurons decreased significantly in DCG Ⅳ groups after 7 and 14 times infrasound exposure. Moreover, in the 7 times and 14 times infrasonic exposure groups, the densities of the injured neurons obviously decreased in cortex and edema in brain was alleviated with DCG IV intervention. CONCLUSION Our study indicates that DCG Ⅳ, agonist of groupⅡ mGluRs, might be used to attenuate the injury of cortex neurons induced by infrasound.
