调肝导浊中药对高脂血症小鼠巨噬细胞集落刺激因子mRNA表达影响的实验研究

摘　　要：目的:观察高脂血清对小鼠体外培养腹腔巨噬细胞(MΦ)、巨噬细胞集落刺激因子(M-CSF)mRNA表达的影响,探讨调肝导浊中药的调控作用及防治动脉粥样硬化(AS)的机理所在。方法:制备小鼠腹腔MΦ,提取RNA,采用反转录聚合酶链反应(RT-PCR)技术观察M-CSFmRNA的表达。结果:高脂组M-CSFmRNA表达显著增强(与正常组比较P<0.01);中药组M-CSFmRNA表达较模型组减弱,较正常组增强。结论:调肝导浊中药对M-CSFmRNA表达具有调控作用,这一调控机制对AS起到良好的防治作用。Objective:To observe the influence of the high lipid serum on expression of the peritoneal M-CSF(macrophage-colony stimulating factor)mRNA in mice M?and to study the mudulating effect of Chinese herbs which can regulating the liver and removing the turbidity and its mechanism of preventing and treating AS(arteriosclerosis).Methods :M?in the abdominal cavity fluid was harvested .Then RNA was obtained.The expression of M-CSF mRNA was observed with RT-PC R.Results:The expression of M-CSF mRNA in the high lipid group was enhanced obviously(in contrast with the normal group P<0.01).The expression in th e Chinese herbs group was lower than in the model group,but higher than that i n the normal group.Conclusion:The Chinese herbs,which can regulate the liver and removing the turbidity can control the expression of M-CSF mRNA and has a good effect on preventing and treating AS through this mechanism.
