摘 要:目的 探讨腺苷预处理对缺血 再灌注心肌细胞膜损伤的保护作用。方法 将培养 5天的SD乳鼠心肌细胞随机分成 4组 :正常组 :常规条件下 (DMEM培养基及 95 %空气 +5 %CO2 气体环境 )培养 5 0min ;拟缺血 /再灌注组 :先在缺糖缺氧条件下 (无糖Eagle培养基及 95 %N2 +5 %CO2 气体环境 )培养 30min ,再恢复常规条件培养 2 0min ;拟缺血预处理组 :先缺糖缺氧培养 5min ,再复氧复糖培养 5min ,反复 3次后按模拟缺血 /再灌注组操作 ;腺苷预处理组 :用含腺苷 (0 .15 g/L)培养液在常规条件下培养 10min ,再作拟缺血 /再灌注组处理。扫描电镜下观察各组细胞的变化 ,并用胶体苯胺染色及Ridit分析法对PLA活性进行半定量。结果 与正常组比较 ,模拟缺血 /再灌注组细胞的质膜和线粒体的结构损伤严重 ,PLA活性显著性增强。而模拟缺血预处理和腺苷预处理组细胞质膜和线粒体损伤较轻 ,PLA活性显著低于拟缺血 /再灌注组。结论 腺苷预处理对“缺血 再灌注”损伤心肌的细胞膜有保护作用 ,其机制可能与腺苷直接或间接地抑制PLA活性 ,增强质膜的稳定性有关。? Objective To explore the protective effect of adenosine preconditioning on ischemia/reperfusion-induced membrane injury of myocardial cells. Method Enzyme isolated cardiac myocytes from neonatal SD rats were cultured for 5 days and then randomly divided into four groups. Normal group: the cells were cultured for 50-min in normal conditions (DMEM in 95% air + 5% CO 2). Mimic ischemia/reperfusion group: the cells underwent a 30-min 'ischemia' ( in Eagle medium without glucose, 95% N 2+ 5% CO 2) and then a 20-min 'reperfusion' in normal conditions). Mimic ischemic preconditioning group: the cells were preconditioned by 3 times of 5-min'ischemia' followed by a 5-min interval in normal conditions, and then underwent 'ischemia/reperfusion' as above.Adenosine preconditioning group: the cells were preconditioned by a 10-min culture in 0.15g/L adenosine in normal conditions before undergoing 'ischemia/reperfusion'. The ultrastructure of the cells in all groups was examined with scanning electron microscope. The activity of phospholipase A (PLA) was semi-quantitated with colloid aniline staining method and Ridit analysis. Result Compared with the normal group, the cell membranes in the mimic ischemia/reperfusion group were seriously damaged, with a significantly increased activity of PLA. However, the cell membranes in both the mimic ischemic and adenosine preconditioning groups were only slightly injured, and the activity of PLA was significantly lower than that in the mimic ischemia/reperfusion group. Conclusion Adenosine preconditioning has a protective effect on the ischemia/reperfusion-induced membrane injury of myocardial cells, which is probably due to improved membrane stability through the direct or indirect inhibition of PLA by adenosine. 〔
