摘  要:目的 观察单纯疱疹病毒 1型 (HSV 1)感染对体外原代培养鸡胚端脑神经元活性的影响。方法 在Vero细胞 (非洲绿猴肾细胞 )上接种HSV 1,并利用空斑实验测定病毒滴度 ;以 10 6 PFU ml病毒吸附感染原代培养鸡胚端脑神经元 ,用倒置显微镜、透射电镜、MTT细胞活性分析、DNA琼脂糖凝胶电泳 ,观察HSV 1感染对细胞活性的影响。结果 HSV 1感染神经元 2 4 ,4 8,72h与同时正常细胞比较 ,活性分别下降 2 5 % ,5 6 % ,97%。形态学及DNA电泳显示神经元主要以坏死的形式死亡。结论 HSV 1感染导致神经元活性下降。Objective To observe the effect of HSV 1 infection on the viability of the cultured chicken embryo telencephalon neurons in vitro .Methods Primary culture model of chicken embryo of telencephalon neurons was established.Herpes simplex virus type 1(HSV 1) was propagated in Vero cells and viral titer was measured by plaque forming method.Inverted microscope, transmission electron microscope,MTT cell viability and DNA agarose gels electrophoresis were used to evaluate the effect of HSV 1 infection on the neurons.Results At 24,48,72 hours post infection the viability of the neurons decreased by 25%,56%,97% as compared with the control group.Morphological changes and DNA agarose gel electrophoresis showed a necrotic effect.Conclusion HSV 1 infection induced a remarkable decreas on the viability of neurons by necrosis rather than apoptosis.
