兔骨髓细胞成心肌细胞体外分化条件的研究

摘　　要：目的　研究兔骨髓细胞体外分化为心肌细胞的条件。方法　无菌条件下抽取兔胫骨骨髓进行培养。于培养第 3d加入诱导剂 5 氮胞苷 10 μmol/L ,观察 14d。采用免疫细胞化学方法检测被诱导细胞心肌特异性肌钙蛋白 T的表达。结果　 5 氮胞苷诱导的细胞形态与心肌细胞形态类似 ,且心肌特异肌钙蛋白 T染色阳性 ,阳性率为 33%。结论　兔骨髓细胞在 5 氮胞苷诱导下可分化为心肌细胞 ,为今后自体细胞移植提供了良好的细胞来源。Objective To investigate the factors governing the differentiation of bone marrow cells in vitro.Methods Rabbit bone marrow cells were aspirated from tibia,and the cells were incubated with 10μmol/L 5-azacytidine on the third day for 24 hours.At the end of the fourteenth day,cardiac specific protein Troponin T was detected through immunocytochemistry.Results Morphology of cells induced by 5-azacytidine was similar to that of cardiomyocytes.In addition,the cells stained positive for cardiac Troponin T,the positive rate was 33%.Conclusions Rabbit bone marrow cells can be induced into cardiomyocytes by 5-azacytidine,which provides better sources for autologous cell transplantation.
