摘 要:目的探讨倒卵叶五加总皂甙 (SAOH)的体内外抗氧化作用。方法在体实验采用结扎左冠状动脉前降支 30min、再灌注 4 0min复制大鼠心肌缺血再灌注损伤 (IRI)的模型 ,结扎前 1 0min分别舌下静脉注射SAOH5 0mg/kg、1 0 0mg/kg ,观察SAOH对再灌注产生氧自由基 (OFR)的作用 ;体外实验采用抗坏血酸体系产生羟自由基 (·OH- )的原理 ,观察SAOH清除·OH的作用。结果SAOH能提高心肌IRI后血浆超氧化物歧化酶 (SOD)活性 ,减少组织丙二醛 (MDA)的生成量 ;减轻IRI后心肌细胞琥珀酸脱氢酶 (SDH)及细胞色素氧化酶 (CCO)的损伤性反应。SAOH浓度在 0 .2~ 1 .4g/L时体外·OH- 清除率为38.5 %~ 1 0 0 % ,且呈量效关系 (r=0 .91 )。结论SAOH具有抗氧化作用 ,其机制与清除·OH- 有关。Objective To study the effects of Saponin of Acanthopanax Obovatus Hoo (SAOH) on anti-oxidation in vivo and vitro.Methods Myocardial ischemia-reperfusion injury (IRI) model was established by left descending anterior coronary artery (LAD) ligation in 30 min,followed by 40 min of reperfusion.Pretreat with 50mg/kg and 100mg/kg of SAOH,and observe the effects of SAOH on oxygen free radical (OFR) induced by reperfusion injury in vivo;hydroxyl free radical (·OH -) was induced by ascorbate-Cu 2+ reaction system,to study the effect of SAOH on scavenging·OH - by colorimetry in vitro.Results SAOH could increased the ability of superoxide dismutase (SOD) and reduced contents of the malondialdehyde (MDA);SAOH could obviously alleviated damage of succinate dehydrogenase (SDH) and cytochrome oxidase (CCO) induced by reperfusion injury.SAOH from 0.2 g/L to 1.4 g/L showed marked effects on scavenging hydroxyl radical from 38.5~100%,and there was a dose-effect relationship (r=0.91).Conclusion SAOH have anti-oxidation effects,and the effects may be associated with scavenging hydroxyl free radical.
