摘 要:目的 :已有的研究表明HWAP I对大鼠的生理痛、急性和亚急性病理痛均有良好的镇痛效应。本文以吗啡作为阳性对照 ,主要研究了硬膜外注射HWAP I对大鼠慢性神经病理痛的镇痛作用。方法 :本实验以不同内径的单套束缚大鼠单侧坐骨神经造成慢性神经病理性痛模型 ,该模型与慢性束缚性损伤模型 (ChronicConstrictionInjuryModel,CCI模型 )极为相似。对术后第五天的大鼠从其硬膜外注入不同剂量的HWAP I、盐酸吗啡 ,运用光热辐射测痛和非伤害性机械刺激测痛两种测痛方法比较了二者给药后 30、6 0和 12 0分钟的作用效果。结果 :2 0 μg/kg、4 0 μg/kgHWAP I对模型侧足的热痛过敏反应和机械触诱发痛觉超敏反应均有明显的镇痛作用 ,并维持 2个小时以上 ;盐酸吗啡 4 0 μg/kg对大鼠双侧脚的热痛过敏产生即时而短暂的阻断效应 ,对机械触诱发痛觉超敏无影响。结论 :硬膜外注入HWAP I能有效的缓解坐骨神经束缚性损伤大鼠的神经病理性痛。Objective: The previous research indicated that HWAP I has analgesic effect in rat on physiological pain, acute and subacute pain. The present study was undertaken to compare antinociceptive properties of HWAP I and morphine in rats with chronic neuropathic pain. Methods: Chronic neuropathic pain was produced by placing a single polyethylene tube of varied ID around the rat's sciatic nerve, and the model is very similar to the CCI model. Five days after the surgery, different doses of HWAP I and morphine were administered epidurally respectively. Radiant heat method and tactile mechanical stimulating method were used to acesses the analgesic effects 30, 60, 120 minutes after the administration. Results: 20 μg/kg and 40 μg/kg HWAP I blocked the established heat hyperalgesia and mechanical allodynia in the injured hindpaw. Injection of 40 μg/kg morphine produced immediate and short lasting blockade effect on heat hyperalgesia bilaterally but not the allodynia. Conclusion: Epdurally administered HWAP I could allay the rat's sciatic nerve cuff induced neuropathic pain.
