精氨酸对小鼠创伤后免疫反应的影响

摘  要:作者采用双后肢截肢小鼠作为创伤模型,观察创伤后免疫功能的变化和精氨酸的免疫调节作用,以及两者与血浆三碘甲状腺原氨酸(T_3)水平变化的关系.结果表明,截肢小鼠盲肠结扎穿刺(CLP)后的死亡率(73.7%)较对照组(38.2%)明显升高(P<0.05).小鼠截肢后48h脾淋巴细胞对PHA刺激的增殖反应能力受到了明显抑制(P<0.01).从术前48h起,腹腔内注射盐酸精氨酸(0.75mg/g体重,每日2次),能够明显降低创伤小鼠CLP后的死亡率(73.7%:23.1%,P<0.05),并可改善受到抑制的脾淋巴细胞转化功能(P<0.05).血浆T_3水平在术后第1,3日截肢组(1.347±0.207 ng/ml,1.635±0.347 ng/ml)和截肢精氨酸组(1.265±0.376ng/ml,1.245±0.311 ng/ml),均较正常组(3.042±0.611 ng/ml)明显降低(P<0.01),术后第3日截肢精氨酸组降低特别明显.由此可见,精氨酸对创伤后机体的免疫增强作用与T_3无关.For the purpose of examining the changes of the immune system along with the immunomodulating effect of argjnine and their relationships to the change of plasma iT3 after trauma, mice with their bilateral hindlimbs amputated were used as trauma models. The results indicated that the mortality of traumatized mice after cecal ligation and puncture (CLP) (73.7%) was significantly higher than that of the controls (38-2%, P<0.05). The PHA 'induced spleen lymphocyte proliferation was markedly depressed 48 h after amputation (P<0.01). Intraperitoneal irjection of argjnine (0.75 mg/g body weight, 2/d) started 48 h before amputation could decrease the mortality of traumatized mice after CLP (23.1%, P<0.05) and improve depressed T cells transforming function (P<0.05). The plasma T3 level of amputated mice (1.347±0.207 ng/ml; 1.635±0.347 ng/ml) and trauma+arginine mice (1.265±0.376 ng/ml; 1.245±0.311 ng/ml) on the 1st and 3rd day are all lower than that of the normal group (3.042±0.611 ng/ml )(P<0.01), particularly the trauma±arginine group on the 3rd day. Therefore, there is no relationship between the action of arginine on the immune system after trauma and the plasma level of T3.
