摘  要:为了确定轮叶党参RAPD分析的最佳PCR条件进行了各种反应条件实验.结果表明:PCR反应时,总反应液20μL中基因组DNA浓度为20ng,引物浓度为10pmol,退火温度为39.1℃和41.3℃,扩增次数在40次下出现可辨认的鲜明的谱带.由此,在上述条件下PCR反应的结果,49种引物中,用轮叶党参基因组DNA能够形成PCR产物并可确认的引物有20个,形成的总谱带数为77个,GC含量高(平均66.5%)形成的谱带也好.The effect of several paramenters on PCR amplification for RAPD in condonopsis lanceolata were studied.The results showed that bands were clear in following conditions: Approximately 20ng of genomic DNA volume and 10 pmol primers;annealing temperature was 39.1 ℃ or 41.3 ℃;40 cycles. In results of using above PCR condition,primers that formed polymorphic bands from genomic DNA of codonopsis lanceolata among 49 primers were 20,and formed 77 number of bands, there were appeared well as the higher(average 66.5%) GC content.
