摘 要:将离体分化的非洲菊小茎芽接种在不含碳源的培养基上,于4mg/m3CO2、2800lx日光灯照的条件下强迫其自养生长,并与异养生长的离体培养物进行对比试验。结果表明,异养状态的小茎芽生根率、叶面积及干重(100%、2.78cm2·株-1、51.4mg·株-1)明显地高于自养状态的小茎芽(22.6%、1.04cm2·株-1、35.4mg·株-1),其再生的小植株也比后者强壮;在自养培养的条件下进一步提高光照强度或补加外源的碳源对根的分化和营养体的生长未见明显的影响。 Gerber jamesonii shootbud propagules were cultured on the sugar containing or sugar free media with or without enhanced CO2 concentration ( 4 mg/m3 )in the chamber around the airtight vessels or ventilated vessels with a gas permeable film. The rate of morphogenesis in term of root differentiation rate and leaf number and the growth in term of the leaf area and dry weight of plantlet in ventilated vessels with surrounding CO2 riched atmosphere were higher than those in conventional cultural conditions. The morphogenesis rate and growth of plantlets formed on sugar containing media in containers with gas permeable film closure were greater than those on sugar free media. The same differences were not measured between the sugar free and containing media in airtight vessels. The results suggest that using sugar free medium and ventilation vessel with CO2 enriched environment can indeed force Gerber jamesonii shootcuttings cultured in vitro from exotrophic to autotrophic and improve their growth and root differentiation. The plantlets produced in these conditions are acclimatization and of better growth.
