良园枇杷膏对豚鼠气道收缩和炎症反应的作用

摘　　要：目的 :良园枇杷膏 (下简称枇杷膏 )是一个传统的镇咳祛痰复方中成药。本文旨在研究其对气道平滑肌的扩张和抗肺部过敏性炎症的作用。方法 :采用肺功能测定法、离体气管法观察枇杷膏的支气管扩张和抗过敏作用 ;支气管肺泡灌流法观察抗炎作用。结果 :在整体试验中 ,枇杷膏 7.5 g/kg和 15 .0 g/kg i.g抑制致敏豚鼠抗原攻击后引起的肺阻力 (RL)增加和动态肺顺应性 (Cdyn)降低 ;抑制支气管肺泡灌流中嗜酸性粒细胞增加 ,ID50 (95 %可信限 )为 1.98(0 .41～ 9.64 ) g/kg;在离体试验中 ,枇杷膏呈剂量依赖反应松弛豚鼠离体气管平滑肌 ,IC50 (95 %可信限 )为 1.2 3 (0 .97～ 1.5 4)× 10 -4 g.L-1 ;抑制 Schultz-Dale反应 ,ID50 (95 %可信限 )为 4.5 5 (2 .78～ 7.42 )× 10 -5g.L-1。结论OBJECTIVE:Liangyuan Pipagao ( LYPPG )is a compound preparation of traditional Chinese medicine applied to antitussive and expectorant. The effect of the drug on brochoconstriction and inflammation in the sensitized guinea pig were evaluated. METHOD:pulmonary mechanics changes in the ovalbumin sensitized guniea pigs induced by ovalbumin aerosol in vivo, Schultz Dale reaction of the ovalbumin sensitized trachea and isolated resting tracheal relaxing in guinea pigs in vitro were investigated. RESULTS:LYPPG 7.5 g/kg and 15.0 g/kg by ig remarkably prevented the alteration of lung resistance( R L ) and dynamic lung compliance ( C dyn ) induced by antigen challenge, and 0.83g/kg,2.5g/kg and 7.5g/kg by ig inhibited antigen induced inerense of eosinophils in the bronchoalveolar lavage fluid of the sensitized guinea pigs in vivo, ID 50 value (95% confidence limits)= 1.98(0.41～9.64)g/kg. LYPPG relaxed the rest isolated tracheal smooth muscle in dose dependent manner, IC 50 (95% confidence limits)= 1.23 (0.97～1.54)×10 4 g.L 1 ,and inhibited Schultz Dale reaction in guinea pigs in vitro, ID 50 (95% confidence limits)= 4.55(2.78～7.42)×10 5 g.L 1 . CONCLUSION:These results indicated that LYPPG had a significant pharmacological effects on bronchodilatation and anti allergy inflammation.
