摘  要:目的 改进差示分光光度法测定血清中苯巴比妥 (PB)的浓度。方法 在酸性条件下 ,以三氯甲烷提取2次 ,利用PB在碱性、酸性介质中吸光度差值建立标准曲线 ,按差示分光光度法测定血清中PB的浓度。结果 血清PB浓度线性范围 2~ 80 μg·ml-1,平均回收率为 10 0 .6 6 %± 0 .36 % ,日内RSD =2 .17% ,日间RSD =2 .5 9% ,血清PB在pH 10条件下 ,2 5min内较稳定 ,抗干扰性良好。结论 所用方法简便、快捷、灵敏度高、重复性好 ,所需的设备及试剂经济易得 ,适于中小医院血药浓度检测。OBJECTIVE To optimize differential spectrophotometry for the determination of phenobarbital PB in serum.METHODS PB in serum was extracted two times with trich oromethane under acidic condition. The standard curve of PB in serum was established according to the absorbance difference of PB in basic and acidic media, and the PB in serum was determined by differential spectrophotometry. RESULTS The linear range of PB in serum was from 2 μg·ml -1 to 80 μg·ml -1 , The average recovery was 100.66% ±0.36% with RSD 2.17% and 2.59 % for within a day and between days. The PB in serum was stable within 25 min under base conditions of pH 10.CONCLUSION The method is quick,simple, high sensitive and well reproducibile. The instrument and reagents were cheap and easy to obtain.It is suitable for clinical drug monitoring in hospital of small or middle scale.
