摘  要:目的:探讨大鼠外周血白细胞介素1β(IL-β)和下丘脑室旁核中的一氧化氮是否具有昼夜节律以及两者的相关性。材料和方法:成年雄性SD大鼠24只随机分为4组,分别在16:00、22:00、4:00和10:00时腹腔注射细菌脂多糖。两小时后,分别麻醉采血及灌注取脑。采用放射免疫分析法测定外周血IL-1β的含量,运用β-NADPH-酶组化和计算机图像分析技术对室旁核中NADPH-d阳性神经元的切面面积和平均灰度值进行分析。结果:1.6:00时组外周血IL-1β含量最高,12:00时组最低,显示出明显的昼夜节律性。2.四组大鼠室旁核内NADPH-d阳性神经元的切面面积12:00时组最大,18:00时组最小,差异有显著性。3.外周血IL-1β含量昼夜节律变化的时相比室旁核一氧化氮产生量昼夜节律变化的时相早6小时。结论:大鼠外周血中IL-1β和室旁核一氧化氮的产生量均有昼夜节律性变化,外周血中IL-1β可能对室旁核一氧化氮的产生有促进作用。Objective:To study circadian rhythm changes and the relation between the nitric oxide(NO)in the hypothalam-ic paraventricular nucleus(PVN) and interleukin 1β(IL-β)in the peripheral blood in rats. Methods; A total of 24 adult male SD rats were randomly divided into 4 groups according to intraperitoneal injection with lipopolysaccharide(LPS)at 16 :00,22:00: , 4:00 and 10:00 oclock respectively. Two hours after inj ection, the rats were anesthetized and their peripheral blood were taken, then the animal were perfused and their brain were taken. The levels of IL-1β in the peripheral blood were measursed using radioactive analysis, and the sectional area of NOS-positive neurons in the PVN measured with NADPH diaphorse histochemistry and microcomputerized image analysis. Results:(1)The peaks of IL-1β level were at 6: 00h,and reached the lowest level at 12-00. The levels of the IL-1β in the peripheral blood showed significant circadian rhythm. (2)The biggest sectional area of NOS positive neurons in the PVN was at 12:00h,and reached the smallest at 18 :00. Significant difference of the NO levels in the PVN between these 2 groups was found. (3)The circadian rhythm phase of IL-1β level in the peripheral blood was 6h earlier than that of the NO production in the PVN. Conclusion peripheral blood IL-1β level and PVN NO levels show has significant circadian rhythm. IL-lfi in the peripheral blood may enhance the productions of NO in the PVN.
