双亲型吸附剂对HDL的吸附动力学研究

摘  要:设计合成了以葡聚糖为载体,同时具有亲水性磺酸基和疏水性胆固醇两类配基的新型吸附剂。通过对高密度脂蛋白 (HDL) 纯溶液中吸附等温线的测试,比较了以Dextran G-75为载体双亲型吸附剂与单一亲水型磺酸基配基、单一疏水型胆固醇配基吸附剂吸附量和亲和吸附系数的关系,对双亲型吸附剂的吸附动力学进行了初步研究。在HDL溶液中,亲水型磺酸基、疏水型胆固醇配基、双亲型吸附剂对HDL的吸附曲线基本符合Langmuir吸附方程。In order to study the adsorption kinetics, hydrophilic dextran sulfate adsorbent (DS), hydrophobic cholesterol modified dextran adsorbent (CMD) and a new type amphiphilic dextran adsorbent were synthesized. Kinetics studies were made for hydrophilic, hydrophobic and amphiphilic adsorbents. The adsorption isotherms of the three adsorbents at 37℃ were investigated after measuring the adsorption capacity. Experimental results show that the shapes of these three curves are similar to that of Langmuir type. The maximal HDL adsorption capacity Q∝(mg/ml gel) are 0.055 (DS), 0.117 (CMD) and 0.220 (AD) respectively. The relative avidity coefficients (Ka) of different adsorbents are 13.78 (DS), 38.99 (CMD) and 92.97 (AD) respectively. Furthermore, while interacting with amphiphilic HDL adsorbent, the HDL seems to be adsorbed by different mechanisms that include hydrophilic ionic adsorption and hydrophobic adsorption. From the experimental data and the result of elemental analysis, we found that synergistic effect existed between the two types of ligands of amphiphilic adsorbent.
