摘 要:目的:建立用HPLC方法测定注射洛美沙星后不同时间的胆汁药物浓度。方法:胆汁样品离心后取上清液直接进样分析。色谱条件为C18柱(250 mm×4.6 mm,5μm),流动相:ψ(0.05 mol/L柠檬酸液∶1 mol/L乙酸铵溶液∶乙腈)=77∶1∶14,流速:1.0mL/min,检测波长:287 nm。结果:胆汁杂质对测定影响不大,线性方程相关系数大于0.999,平均回收率为85.28%。结论:方法专属性好,适用于临床胆汁中洛美沙星浓度测定。Objective: To establish a HPLC method for the determination of the drug concentration in bile after the injection of lomefloxacine. Methods: The bile sample was centrifuged and the supernatant was used in the determination. The mobile phase consisted of 0. 05 mol/L citricacid - 1 mol/L Ammonium acetate - acetonitrile(77 : 1 : 14) . The YWC18 (250 nm×4. 6 mm,5μm) column was used, with the velocity of flow at 1. 0 mL/min and the detection wavelength of 287 nm. Results: The correlation coefficient of the linear equation was larger than 0. 999 ,the average recovery was 85. 28% . Conclusion: The method was simple, rapid and specific. It can be used for determination of Lomefloxacine in bile.
