左旋四氢巴马汀对大鼠催乳素水平低下模型的拮抗作用

摘　　要：研究左旋四氢巴马汀 (L tetrahydropalmatine ,L THP)对哺乳期催乳素 (Prolactin ,PRL)水平低下模型大鼠中枢多巴胺 (Dopamine,DA)D2 受体的拮抗作用。通过对哺乳期大鼠皮下注射阿扑吗啡(Apomorphine,APO) 2 5mg·kg- 1·d- 1和 5 0mg·kg- 1·d- 1,以确定用APO建立哺乳期大鼠PRL水平低下模型的有效剂量。并在此模型基础上研究L THP对APO诱导的哺乳期大鼠PRL水平低下的拮抗作用。结果表明 ,皮下注射APO 5 0mg·kg- 1·d- 1可显著降低哺乳期大鼠血清PRL水平 ,抑制仔鼠体重增长 ,可造成哺乳期大鼠PRL水平低下模型。L THP 6 0mg·kg- 1·d- 1可显著性提高哺乳期大鼠血清PRL水平 ,促进仔鼠体重增长。L THP可能是D2 受体拮抗剂。Objective: To study the effects of L-tetrahydropalmatine,the Dopamine D 2 antagonist,on low prolactin level model induced by Apomorphine of lactational rats.Method: Compared the low prolactin level model established by Apomorphine 2.5mg·kg -1·d -1 s.c.with model by 5.0mg·kg -1·d -1s.c..Study the influence of L-tetrahydropalmatine on milk-secretion by detecting serum prolactin level of lactational rats and weight gain in sulking rats.Results: Apomorphine 5.0mg·kg -1·d -1 s.c.remarkably decreased the prolactin level of lactational rats and made rats in sulking gain weight.L-tetrahydropalmatine 60mg·kg -1·d -1 s.c.remarkably improved prolactin level and gained weight.The effects of L-tetrahydropalmatine 30mg·kg -1·d -1 s.c.were not remarkable.Conclusions: Apomorphine 5.0mg·kg -1·d -1 s.c.could establish low prolactin level model.L-tetrahydropalmatine might be Dopamine D 2 receptor antagonist.
