摘 要:目的 :确立影响抗癌新制剂——白花蛇舌草胶囊生产工艺的主要因素 ,并制定最佳生产工艺。方法 :以正交设计的实验方法 ,对组方中抗癌有效成分丹参酮进行定量分析 ,对其它有效物质进行收率分析。在醇提工艺中 ,选取L8( 2 7)正交表 ,考察溶剂浓度、溶剂稀释倍数、提取次数、提取时间等工艺参数对丹参酮含量的影响。在水煎工艺中 ,选取 L9( 3 4 )正交表 ,考察药材浸泡时间、加水量、提取次数、提取时间等工艺参数对产出率的影响。结果 :方差分析表明 ,在醇提工艺中 ,药材浸泡时间和溶剂稀释倍数对提取丹参酮影响较大 ;在水煎工艺中 ,加水量对提取量影响显著。本文设计交互作用影响均较小。结论 :最佳制备工艺为 :用 6倍、4倍生药量的 95 %乙醇将丹参回流提取两次 ,提取时间各为 2小时和 1小时 ,丹参药渣与其它药材合并 ,然后加 10倍、6倍生药量的水煎煮两次 ,每次 2小时 ,过滤、浓缩、醇沉 ,所得醇沉液浓缩 。Objective: To study the main effective factors of the producing technology of Herba(Hedyofis Oldenlandia diffusa) capsule which is a new drug against cancer. Methods: Determining content of TanshinoneⅡ_A which is considered an effective element of the drug by orthogonal experiment and analyzing weights of the other efficient elements. Chosen Orthogonal table L_8(2 7) in the experiment of extracting in alcohol, to research the effect of content of TanshinoneⅡ_A from technological parameters(example solvent-density, dilution-multiple, extraction times, the time of extraction etc). Chosen Orthogonal table L_9(3 4) in the experiment of decoction technology to research the productivity of TanshinoneⅡ_A from technological parameters(example the time of the herb marinating, the volume of water filled, extraction times, the time of extraction etc). Results: Through the analysis of the variance herb marinating time and diluted concentration has significant effect on the content of tanshinoneⅡ_A in the experiment of extracting in alcohol, and the volume of water filled is a significative parameter in the decoction technology. The effect of interaction in the designed test is tiny. Conclusion: The best producing technology is obtained. Tanshinone was extracted 2 times using 95% alcohol <solvent: herb=6:1(1st) or 4:1(2nd)>whose volume as sextuple and fourfold herb, the time of extraction was 2h and 1h. The mixture residue leavings of Tanshinone and other herb, filled water <water: herb=10:1(1st) and 6:1(2nd)>wae decocted, 2h/time, and filtered, concentrated, precipitated in alcohol, then concentrated the liquid, and mixed with liquid of Tanshinone extracted.
