青蒿素对体外培养的HeLa细胞生长的影响

摘　　要：目的研究青蒿素对体外培养的HeLa细胞生长的抑制作用。方法 用台盼蓝染色观察药物对细胞的增殖抑制率;MTT法测定药物的抗肿瘤活性;透射电镜观察药物对肿瘤细胞形态学的影响;流式细胞仪检测细胞凋亡率。结果 台盼蓝染色法证实青蒿素对HeLa细胞有抑制作用,抑制率与药物浓度正相关;细胞经药物作用48h后的IC50为55.58μmol/L;电镜观察细胞有典型的凋亡形态特征(凋亡小体);流式细胞仪检测细胞凋亡率在一定范围内与药物浓度正相关。结论青蒿素可抑制HeLa细胞的生长,诱导HeLa细胞凋亡。Objective To investigate the effect of artemisinin(ART) on the growth of HeLa cells in vitro. Methods The antiproliferation activities of ART were evaluated on HeLa cells by means of trypan blue dye and MTT assay. The morphological changes of ART on HeLa cells were observed under transmission electromi-croscope. Cell survival ratio influenced by ART was assessed by a flow cytometer. Results After HeLa cells were treated by ART for 48 hours, the growth of the cells were inhibited in vitro , the IC50 of ART was 55.58 umol/L; there were some typical changes of apoptosis by transmission electromicroscopy. Conclusion ART can inhibit the proliferation of HeLa cells and induce their apoptosis.
