摘 要:目的 动态观察安氟醚吸入麻醉大鼠不同时期各脑区Na+ ,K+ ATP酶活性变化 ,探讨安氟醚麻醉作用机制。方法 40只SD雌性大鼠按麻醉不同时期随机均分为 5组 :对照组、诱导期组、麻醉期组、恢复期组和清醒期组。用分光光度法测定不同时期大脑皮层、脑干、海马Na+ ,K+ ATP酶活性。结果 与对照组比较 ,大鼠大脑皮层、脑干、海马Na+ ,K+ ATP酶活性在诱导期即开始下降 ,其中大脑皮层和海马酶活性降低有统计学意义 (P <0 0 5 ) ;麻醉期Na+ ,K+ ATP酶活性下降至最低 (P <0 0 1) ;恢复期酶活性又开始回升 ,但仍低于对照组 (P <0 0 5 ,P <0 0 1) ;清醒期基本恢复至正常水平 (P >0 0 5 )。且麻醉深度变化与Na+ ,K+ ATP酶活性抑制相关。结论 安氟醚可明显抑制Na+ ,K+ ATP酶活性 ,且与行为变化相平行 ,提示安氟醚通过抑制Na+ ,K+ ATP酶活性而发挥麻醉效应 。AIM To observe dynamicly the effects of enflurane on the Na +,K + ATPase activity in different rat brain region at different stage and explore the anaesthetic mechanism of enflurane. METHODS Forty femal rats were divided randomly into five groups as follows: control group; induction group; anaesthetic group; recovery group and awake group. The Na +,K + ATPase activity at different stage in cerebral cortex, brain stem and hippocampus were measured by spectroscopic analysis. RESULTS The activity of Na +,K + ATPase in cerebral cortex,brain stem and hippocampus begun to reduce during the induction stage, especially, the activity of Na +,K + ATPase in cerebral cortex and hippocampus reduced significantly( P <0\^05); it reduced to the lowest during anaesthetic stage and begun to increase during recovery stage but still lower than it in control group( P<0 05,P<0 01) ; during awake stage, it almost recovered to nomal level( P >0 05). Also, the changes of the depth of anesthesia were related to the inhibition of Na +,K + ATPase activity.CONCLUSION Enflurane significantly inhibited the activity of Na +,K + ATPase and it paralled to behavioural changes, which suggested enflurane worked through inhibiting the activity of Na +,K + ATPase, the depth of anaesthesia was related to the degree of inhibition of the Na +,K + ATPase activity.
