摘 要:以高效液相色谱法为定量手段,研究了单诺沙星(danofloxacin)在雏鸡体内的组织动力学特征,并采用回归分析法研究了血药浓度与组织药物浓度间的相关性。60只雏鸡口服单诺沙星(5mg/kg)后,血浆的药时数据符合一级吸收二室模型。肝脏、肾脏、心脏、肺脏的药时数据符合一级吸收三项指数方程;心脏的药时数据符合一级吸收二项指数方程。血浆、肝脏、肾脏、心脏、肺脏及肌肉的主要动力学参数是:t1/2ka分别为0.2428、0.3524、0.2643、0.3377、0.2536h,t1/2a分别为0.8917,1.1212,0.7189,1.16142,0.4437h(缺肌肉的t1/2a),t1/2β分别为8.7936,3.6025,4.4570,10.2940,4.3283,5.9578h,Tp分别为0.9377,1.0665,0.9362,0.9993,0.9962,1.4381h,Cmax分别为0.5487,2.8419,2.4295,0.2668,1.7042,0.2726mg/L,AUC分别为3.0523,13.586,0,10.4182,2.1521,9.6888,2.7157/mg(L·h),Tcp分别为12.6130,0,18.073,0,18.879,0,8.055,7,19.6930,13.8600h。研究结果表明,组织中的药物浓度以肝脏、肾脏最高,其次是肺脏,均显著高于血浆中的药物浓度,心脏、肌肉中的浓度最低,血浆的有效浓度维持时间均显著长于各组织。The tissue pharmacokinetics and residues of dan o floxacin in chickens were investigated by the HPLC method,and the dynamic inter related analysis of danofloxacin levels between plasma and tissue were performed with regression analysis method.Following oral administration 5 mg/kg danofloxa cin the concentration-time course in the plasma could be described by two-comp artment open model with first-order absorption rate.The concentrations-time co urse of the liver,kidney and lung were best described by triexponential equation ;and the muscle performed to bioexponential equation.The main kinetic parameters for the plasma,liver,kidney,heart,lung and muscle in the chickens were as follo wing,respectively:t 1/2ka: 0.242 8, 0.352 4, 0.354 1, 0 .264 3, 0.337 7 and 0.253 6 h. t 1/2α: 0.891 7, 1.121 2, 0.718 9, 1.614 2 and 0.443 7 h. t 1/2β: 8.793 6, 3 .602 5, 4.457 0, 10.294 0, 4.328 3 and 5.957 8 h. Tp: 0. 937 7, 1.066 5, 0.936 2, 0.999 3, 0.996 2 and 1.438 1 h. C max: 0.548 7, 2.841 9, 2.429 5, 0.266 8, 1.704 2 and 0 . 272 6 mg/L. AUC: 3.052 3, 13.586 0, 10.418 0, 2.152 1, 9.68 8 8 and 2.715 7 mg/(L·h). T cp分别? ?2.613 0, 18.07 3 0, 18.879 0, 8.055 7, 19.693 0 and 13.860 0 h. The results o f tissues pharmacokinetic investigation showed that the concentrati ons of danofloxacin were the highest in liver and kidney,higher in lung,the lowe st in heart and muscle.All concentrations in liver,kidney and lung significantly exceeded corresponding levels in plasma.Apart from heart,The duration of therap eutic level of danofloxacin(T cp) in the plasma was significantly longer th an that in the tissues.
