摘　　要：目的　探讨含preS2免疫表位基因质粒DNA疫苗诱导小鼠的免疫应答。方法　质粒转染COS 7细胞 ,用ELISA法测定瞬时表达产物。质粒经碱裂解法大量提取 ,Sepharose 4FF柱层析纯化后 ,直接肌肉注射免疫NIH小鼠。检测小鼠的体液免疫和细胞免疫应答效果。结果　preS2表位多肽和HBsAg高效表达。免疫小鼠血清中检测到高滴度的抗 HBs和抗 preS2。淋转刺激指数SI和IL 2活性 ,质粒DNA疫苗与单纯主蛋白疫苗免疫对照组比较 ,差异有显著意义。Objective To explore the immune response of hepatitis B surface antigen DNA vaccine containing preS2 immune epitope in mice.Methods Transform COS 7 cells with recombinant plasmid and detect instantaneously expressed product by ELISA.Extract the recombinant plasmids in a large quantity by alkali lysis,purify by Sepharose 4FF column chromatography,injected i.m. into NIH mice and detect humoral and cellular immune responses.Results Both preS2 epitope polypeptide and HBsAg were highly expressed.High serum anti HBs and anti preS2 titers were detected in the immunized mice.Both splenic lymphocyte transormation stimulating index(SI) and IL 2 activity of the DNA vaccine were significantly different from those of protein vaccine.Conclusion The hepatitis B surface antigen DNA vaccine containing preS2 immune epitope induced humoral and cellular immune responses in NIH mice.
