摘 要:目的:初步探讨纤连蛋白(fibronectin,Fn)与烟曲霉分生孢子的相互黏附作用,以及孢子相互黏附作用的关系。方法:利用流式细胞仪检测不同浓度的Fn与烟曲霉分生孢子的黏附率及经不同胰蛋白酶处理后分生孢子与Fn的黏附率的变化;通过共聚焦免疫荧光显微镜观察Fn受体与分生孢子的相互作用及表面形态学变化;通过扫描电镜和透射电镜观察分生孢子静止期细胞壁最外层的形态及孢子在生发膨胀过程中形态学的变化。结果:随着Fn浓度的增加,与分生孢子的黏附率也增加;当用胰蛋白酶预先处理过烟曲霉分生孢子后,定量的Fn与烟曲霉分生孢子的黏附率随着胰蛋白酶浓度的增加而降低;含有荧光的Fn的配体-受体均匀分布在烟曲霉分生孢子表面;静止期和生发膨胀期分生孢子细胞的表面和内部均有特征性的变化。结论:Fn与烟曲霉分生孢子的黏附作用有浓度依赖性和饱和性。静止期分生孢子的表面可表达高水平Fn受体;膨胀过程中表面小棘状突起逐渐脱落;胞壁内没有受体存在;蛋白质破坏性试剂可除去分生孢子小棘状突起。Objective:To study the inte ra ction of adhesion between the fibronectin and conidia of A.fumigatus and to pro be into the relationship between the pathogenicity of conidia of A.fumigatus an d their surface receptors.Methods :Flow cytometry was used to determine the ra te of adhesion between different concentrations of fibronectin and the conidia of A.fumigatus,and the change of the rate of adhesion after treatment with di fferent concentrations of trypsin.Adhesion of fibronectin to the surface of co nidia was examined by immunofluorescence confocal microscopy;morphology of rest ing and growing phases of the canidia were examined by scanning electron microsc opy and transmission electron microscopy.Results:Along with the increasing of the concentration of fibronectin,the rate of adhesion to the conidia increased .After treatment of the conidia by trypsin the rate of adhesion decreased.Flu orescence containing fibronectin ligand-receptor evenly distributed on the sur face of the conidia.There were characteristic changes inside and on the surface of the resting and growing conidia.Conclusions:Adhesion of fibronectin to con idia is concentration-dependent and can be saturated.High levels of fibronect in receptors can be expressed on the resting conidia.During swelling the surfac e echinulated layer of conidia is disintegrated,and there are no receptors with in the cell wall.Characteristic echinulations of the conidia can be dismantled by proteolysis.
