摘 要:目的对淋病奈瑟菌隐蔽性质粒B(crypticplasmidB ,cppB)基因用于淋病奈瑟菌稳定L型基因诊断的评价。方法用非高渗液体培养基培养青霉素诱导形成和自发形成的淋病奈瑟菌稳定L型 ,获得纯培养物以特异性cppB基因引物进行聚合酶链反应 (PCR)检测。结果淋病奈瑟菌细菌型可检出cppB基因 ,但不论诱导形成还是自发形成的稳定L型传 5代后纯培养物cppB基因检测均为阴性反应。结论细胞壁缺陷可导致淋病奈瑟菌cppB基因丢失 。Objective To evaluate the usability in detection of cryptic plasmid B (cppB) gene for stable L forms in Neisseria Gonorrhoeae. Methods The stable L forms of N.gonorrhoeae which were induced by penicillin or formed spontaneously in non high osmotic liquid medium were subcultured for five times in the same kind of medium to get pure cultures of stable L form and then the cppB genes of the pure cultures were detected by PCR. Results The cppB gene was found in the bacterial form of N.gonorrhoeae subculture in the non-high osmotic liquid medium, but it was not found in the stable L forms. Conclusion The cell wall deficiency would lead to lose of the cppB gene, fasle negetive results existed in detection of N.gonorrhoeae and would result in omission or fail to diagnosis for gonorrhoeae.
