摘 要:研究了甲基单胞菌 Methylomonas sp.GYJ3发酵培养过程中生长特征和产酶特征 ,结果表明该菌株在生长过程中产酸 ,通过控制发酵过程中 p H可提高细胞的生长密度 ,其生长周期较摇床发酵短 .细胞中甲烷单加氧酶产生于细胞的对数生长期 ,在 70 h以后甲烷单加氧酶活性不再增加 ,最高活性为 3.97nmol环氧丙烷 / min· mg干细胞 .同时研究了细菌生长过程中甲烷的利用 ,结果表明每克甲烷的消耗可产生 0 .69g细胞干重 .考察了 Cu2 +浓度对细胞产生可溶性甲烷单加氧酶和颗粒性甲烷单加氧酶的影响 .并由此建立了 GYJ3菌的连续发酵动力学模型 .The characteristics of the growth of Methylomonas sp.GYJ3 and the production of methane monooxygenase were studied. The production of acid was implied by observing the decline of pH during the growth period. The production of methane monooxygenase activity remained almost constant in the later slow growth phase (after 70 h). The utilization of methane was estimated to be 0.69 g of dry cell/g of methane. Effect of Cu 2+ on production of soluble form and particulate form of methane monooxygenase was also studied. The optimal conditions for production of soluble methane monooxygenase was determined, at that its activity was 1.83 nmol propene oxide/mg of dry cell/min. Therefore, the kinetic modeling of continuous fermentation of Methylomonas sp.GYJ3 was established.
