摘 要:目的 :通过比较妊高征孕妇血清与肿瘤坏死因子 α(TNF α)对血管内皮细胞的影响 ,探讨TNF α在妊高征发病中的作用。方法 :将培养的内皮细胞随机分为 6个组 :1组为正常培养基对照组 ,2组为TNF α对照组 (TNF α的含量为 4 0 0u/mL)。 3组为正常孕妇血清对照组 ,4、 5、 6组分别为培养基中加有轻、中、重度妊高征孕妇血清组 (孕妇血清的含量为 30 %vol/vol)。然后观察各组的生长曲线、细胞DNA含量图像分析、细胞形态、超微结构以及细胞间粘附分子 1(ICAM 1)的表达情况。结果 :① 妊高征孕妇血清及TNF α(40 0u/mL)抑制血管内皮细胞增殖的作用均是从加入这些因素第 3天后开始 ,并且妊高征病情越重 ,抑制血管内皮细胞增殖越明显。 3天内二者均表现为促增殖的作用。② 光镜、电镜观察可看出妊高征孕妇血清及TNF α对血管内皮细胞有损伤作用 (血管内皮细胞拉长 ,微绒毛变短甚至成泡状以及内质网扩张、线粒体凝聚等改变 )。③ 妊高征孕妇血清和TNF α还可使血管内皮细胞ICAM 1表达增强。结论 :妊高征孕妇血清对血管内皮细胞有明显的损伤作用 ,这种损伤作用与孕妇血清中的TNF α可能有关。在妊高征发病中TNF α有重要的作用。Objective:To investigate the role of tumor necrosis factor-α (TNF-α)in the pathogenesis of preeclampsia. Methods: In the experiment, the human umbilical vein endothelial cells HUVECswere divided into six groups: Group 1 (control group), the medium without TNF-α or sera from pregnancy women. Group 2,the medium with TNF-α400u/mL). Group 3, the medium with normal pregnancy women's sera. Group 4~6, the medium with preeclamptic patients'sera (mild, moderate, severe (30%vol/vol). The cell growth curve, image pattern analysis of cell DNA content, morphologic change , ultrastructure and expression of ICAM-1 were observed respectively in every group. Results: TNF-α was able to inhibit the proliferation of cells after treatment of 3 days, but within 3 days,TNF-α could promote cell proliferation. The effect of sera from preeclamptic patients on the HUVECs was similar to that of TNF-α. Light and electron seen microscopically, the HUVECs revealed elongation,dilation of the rough endoplasmic reticula, condensation of the mitochondria and the shortening of the microvilli after exposure to the sera of preeclamptic patients and TNF-α, respectively. Expression of ICAM-1 on the HUVECs was increased after incubation in the sera of preeclamptic patients and TNF-α.Conclusion:The sera of preeclamptic patients can damage the HUVECs significantly and the results suggest that the effects of sera from preeclamptic patients on HUVECs are at least partly associated with TNF-α in the sera.
