P物质受体在急性出血坏死性胰腺炎肺组织中的过度表达

摘　　要：目的　了解神经激肽 1受体 (NK 1R)在急性出血坏死性胰腺炎 (AHNP)大鼠肺组织中的表达 ,探讨该受体的表达与AHNP时肺损害的关系。方法　健康成年Sprague Dawley大鼠随机分为AHNP组 (60只 )和正常对照组 (3 0只 )。正常对照组开腹后只翻动胰腺 ,AHNP组大鼠胰胆管恒速逆行注射 5 %牛磺胆酸钠 (0 0 1ml·kg-1·min-1) ,制成AHNP大鼠模型。应用逆转录聚合酶链反应 (RT PCR)检测肺组织NK 1R的mRNA水平 ,应用Westernblot技术检测NK 1R的蛋白水平 ,以免疫组织化学方法进行NK 1R的组织学定位。结果　与正常对照组相比 ,AHNP肺组织中NK 1RmRNA和蛋白都过度表达。免疫组化检查显示 ,NK 1R的表达主要位于肺泡隔血管内皮细胞表面、部分肺泡Ⅰ、Ⅱ型上皮细胞表面等处。结论　AHNP肺组织中NK 1R的表达水平明显上调 ,扰乱了神经激肽的作用环节 。Objective To investigate the expression of neurokinin 1 receptor (NK 1R)in the lung ofacutehaemorrhagicnecrotizingpancreatitis (AHNP) .TherelationshipbetweenexpressionofNK 1R andlunginjurywasalsoevaluated .Methods 90adultSprague Dawleyrats ,withequalnumberofmaleandfemalewererandomlydividedinto 2 groups :theratsingroupAHNPwereinducedbytheretrogradeintra ductalinfusionof 5 %sodiumtaurocholate (0 0 1ml·kg-1·min-1) .Andtheratsinnormalcontrolgroup receivedlaparotomyonly .Sacrificesweremade 12h ,2 4hand 48hlaterinAHNPandnormalcontrolgroup afterinductionrespectively .Reversetranscriptionpolymerasechainreaction (RT PCR)wasusedtodeter minethemRNAexpressionofNK 1R ,WesternblotanalysiswasusedtodetermineNK 1Rproteinexpres sionlevels,andimmunohistochemistrywasusedtolocalizeexpressionsiteofNK 1R .Results NK 1RmR NAlevelwasenhancedexpressedinthelungofAHNPcomparedwithnormalcontrolgroup .Westernblot analysisrevealedoverexpressionofNK 1RproteinlevelexistedinAHNPgroup .Immunohistochemistryre vealedmoderatetostrongNK 1Rimmunoreactivitywaslocalizedtoalveolarmembrane,Ⅰ ,Ⅱepithelium andpolymorphonuclearleukocytesinthelungofAHNP .Conclusions InAHNP ,overexpressionofNK 1R contributetodisrupteneuropeptidesloopbalance,deterioratelunginjury ,andbeinvolvedinpathophysio logicalchangeinthisdisease .
