摘  要:目的 研究侧脑室给予一氧化氮 (NO)前体L -精氨酸 (L -Arg)对清醒大鼠胃运动及胃电的影响及其作用机制。方法 采用侧脑室微量注射药物、应力传感器引导胃运动及银丝电极同步记录胃电等方法。结果 侧脑室注射 0 .5mg或 1mgL -Arg后胃运动及胃电幅度明显增强 ,胃运动频率无明显变化 ,而静脉注射 0 .5mgL-Arg对胃运动及胃电幅度没有影响 ;事先侧脑室注射 0 .2 5mgNO合酶抑制剂L -硝基精氨酸甲酯 (L -NAME)或切断膈下迷走神经 ,则可完全消除L -Arg对胃运动及胃电的影响 ,但切除腹腔交感神经节后不影响L -Arg的作用。结论 侧脑室给予NO前体物质可使胃运动及胃电增强 ,这种效应系中枢作用 ,并且可能通过迷走神经介导 。Objective To investigate the effects of intracerebroventricularly (i.c.v.) administrated nitric oxide (NO) precursor on gastric motility and electricity in conscious rats and the possible mechanisms. Methods The NO precursor L-arginine (L-Arg) and the NO synthase inhibitor Nω-nitro-L-arginine-methyl-ester (L-NAME) were unilaterally microinjected into the intracerebroventricle of conscious rats, and the antral motility and electricity were monitored using extraluminal strain gauge transducers and silver electrodes respectively. Results The amplitudes of gastric motility and electricity were significantly increased after i.c.v. microinjection of L-arg, but no effect on the frequency of gastric motility was noticed. No marked changes were observed when an equal dose of L-arg was injected iv. I.c.v. microinjection of L-NAME or vagotomy completely prevented L-arg from exerting any effect on gastric motility or electricity, but celiac ganglionectomy did not alter the excitatory effects of L-arg. Conclusion The results suggest that NO precursor applied i.c.v. can increase the gastric motility and electricity, and this central effect seems to be mediated by the vagus nerves, while the sympathetic nerves may not be involved in the pathway.
