摘 要:目的 观察不同剂量的氯胺酮对大鼠丘脑一氧化氮合酶 (NOS)和一氧化氮 (NO)含量的影响。方法 SpragueDawley(SD)大鼠 32只 ,雌雄不限 ,体重 2 0 0~ 30 0 g。随机分为四组 (n =8) :组Ⅰ为对照组 ,给予生理盐水 10ml/kg腹腔注射 ;组Ⅱ、组Ⅲ和组Ⅳ分别给予氯胺酮 35ml/kg、10 0ml/kg和 2 0 0ml/kg。组Ⅰ和组Ⅱ在用药后 5min后断头取材 ,组Ⅲ和组Ⅳ在大鼠翻正反射消失后立即断头取材。在低温操作箱内生理盐水冰面上取脑 ,以分光光度法测定NOS活性和NO量 ,Lawry法测蛋白含量。 结果 大鼠腹腔注射氯胺酮35ml/kg后 ,其丘脑的NOS活性和NO量与对照组相比均无明显变化 (P >0 0 5 )而腹腔注射氯胺酮 10 0ml/kg和 2 0 0ml/kg后 ,NOS活性明显降低 ,分别较对照组降低了 4 4 3%和 4 0 % (P <0 0 5 )和 (P <0 0 1) ,NO量也明显减少 ,分别较对照组减少了 4 2 9%和 4 7 1% (P <0 0 5 )或 (P <0 0 1) ;且这两组的NOS活性和NO量也明显低于氯胺酮 35ml/kg组 ,NOS活性分别降低了 4 7 3%和 4 3 2 % (P <0 0 1) ,NOS量降低了4 7 4 %和 5 1 3% (P <0 0 1) ;此两组之间相比较 ,丘脑的NOS活性和NO量无统计学意义 (P >0 0 5 )。Objective To investigate the effect of diff e rent dose of ketamine on nitric oxide synthase (NOS)activity and nitric oxide(NO )output in rat thalums.Methods Thirty-two SD rats were divided randomly into four groups:group1(control group)w erd administered introperitonealy (ip)normal saline 10ml·kg -1 ,the others were administered ketamine 35 mg·kg -1 (group 2),100mg·kg -1 (group 3),an d 200mg·kg -1 (group 4)respectively.Five minutes later,the rats in group 1 and group 2 were decapitated,Those rats in group 3 and group 4 were decapitated at the time of losing righting reflex.The thalums were rapidly dissected from br a in on the saline ice.NOS activity and NO content were measured by spectrophotome try.Results The NOS activity and NO content had no changes afte r ketamine 35 mg·kg -1 ip.After ketamine 100mg·kg -1 and 200mg·kg -1 were administered ip,the NOS activity were decreased by 44 3%and 40%(P<0 05 or P<0 01 vs control group),and the NO con tent were reduced by 42 9% and 47 1% respectively (P<0 01). Compared with group 2,the NOS activity ahd the NO content were much lower too.th e NOS activity were decreased by 47 3% and 43 2% (P<0 01),an d the NO content were reduced by 47 4%and 53 2% respectively(P <0 01).The NOS activity and the NO content had no significant difference in th ese two groups.Conclusion NO may be a important role in the mec hanism of ketamine aneathesia.
