摘 要:目的 :比较研究三黄降糖方和达美康对糖尿病性心肌病变的作用及心肌GluT4mRNA表达的影响。方法 :以STZ造成大鼠糖尿病模型 ,灌胃三黄降糖方和达美康 8周后 ,检测血糖、胰岛素、血脂、心脏重量 ,以RT PCR法测定心肌GluT4mRNA。结果 :糖尿病大鼠心脏重量增加 ,血糖升高 ,血清胰岛素水平降低 ,血清TC、TG、LDL ch升高 ,心肌GluT4mRNA表达降低。三黄降糖方和达美康均可使糖尿病大鼠心脏重量减轻 ,两药对血糖有轻微降低作用 ,而对血清胰岛素水平无明显影响 ;两药均可防止心肌GluT4mRNA降低。达美康还可降低血清TC和LDL ch。结论 :三黄降糖方和达美康对糖尿病性心肌病变具有对抗作用 ,其作用与促进心肌局部GluT4mRNA表达 ,改善心肌细胞能量代谢有关。达美康还可能通过降低血脂而起作用。Objective: To compare the effects of Sanhuangjiangtang decoction (三黄降糖方) and gliclazide on the cardiomyopathy and mRNA expression of glucose transporter 4 (GluT4) in cardiac muscle in diabetic rats. Method: Diabetic model were made by intraperitoneally administered stretozocin in rats. After sanhuangjiangtang decoction and gliclazide were administered orally for 8 weeks in rats, the glucose, insulin and lipid in the blood were determinated, weight of the hearts in rats were weighed, mRNA expression of GluT4 were determinated by RT PCR determination. Results: Weight of the hearts in diabetic rats increased, the blood sugar contents increased, insulin level decreased, total cholesterol, triglyceride and low density lipoprotein cholesterol contents increased in serum, mRNA expression of GluT4 decreased in diabetic rats. Sanhuangjiangtang decoction and gliclazide could reduce weight of the hearts in diabetic rats, lower slightly the blood sugar contents, but serum insulin level had no change in two drug groups. Sanhuangjiangtang decoction and gliclazide conld inhibit decrease of mRNA expression of GluT4. Gliclazide could lower total cholesterol and low density lipoprotein cholesterol contents in the serum in diabetic rats. Conclusion: Sanhuangjiangtang decoction and gliclazide could antagonize diabetic cardiomyopathy, the action mechanism of the drugs may be related to promote mRNA expression of GluT4 in cardiac muscle and improve energy metabolism in cardiac muscle cells in diabetic rats. Gluclazide may be related to lower the blood lipid in diabetic rats.
