摘 要:目的 :比较 HPL C法和微生物法测定血浆中哌拉西林浓度的差异。方法 :6名健康志愿者单剂量静脉滴注哌拉西林 4g,分别用 HPL C法和微生物法测定血浆中药物浓度。 结果 :HPL C法线性范围为 0 .5~ 5 0 0μg/ m l,回收率为 97.40 %~10 0 .0 6 % ,日内、日间 RSD分别为 2 .5 8%~ 3.2 4%和 3.32 %~ 5 .5 9% ;微生物法线性范围为 0 .5~ 10μg/ ml,回收率为98.38%~ 10 6 .85 % ,日内、日间 RSD分别为 3.90 %~ 6 .37%和 7.0 7%~ 9.78%。 结论 :两种方法回收率和精密度均符合要求 ,受试者各时间点平均血药浓度无显著性差异。Objective: To compare the high performance liquid chromatography (HPLC) and microbiological assay for determination of piperacillin in human plasma. Methods: Six healthy volunteers were given a single dose of piperacillin 4 g by infusion. The methods of HPLC and microbiological assay were used to determine the piperacillin plasma concentrations, respectively. Results: The calibration curve of HPLC method was linear in the range of 0.5-500 μg/ml. The recovery rate of HPLC method was 97.40%-100.06%, within-day and day-to-day RSD were 2.58%-3.24% and 3.32%-5.59%, respec-tively. The calibration curve of microbiological assay was linear in the range of 0.5-10 μg/ml. The recovery rate of microbiological assay was 98.38%-106.85%,within-day and day-to-day RSD were 3.90%-6.37% and 7.07%-9.78%,respectively. Conclusion: The recovery rate and the precision of 2 methods are found to be similar. There is no significant difference in piperacillin plasma concentrations measured by HPLC method and microbiological assay.
