摘　　要：目的 :研究肾衰冲剂对大鼠残余肾转化生长因子 β1(TGF β1)、组织金属蛋白酶抑制剂 1(TIMP 1)mRNA过度表达的抑制作用。方法 :制作大鼠 5 / 6肾切除肾衰模型 ,分别予肾衰冲剂及党参、丹参、制大黄灌胃治疗 1个月后取各组大鼠残余肾。用逆转录多聚酶链反应 (RT PCR)法检测残余肾TGF β1、TIMP 1mRNA的表达。结果 :大鼠慢性肾衰模型组表现为TGF β1、TIMP 1mRNA的过度表达 ;经肾衰冲剂治疗后TGF β1、TIMP 1mRNA的表达降低 (P <0 0 1)。其组成单味药丹参、制大黄则有不同程度的作用 ,党参作用不明显。结论 :肾衰冲剂减轻残余肾纤维化的病变与其抑制残余肾组织TGF β1、TIMP 1mRNA的过度表达有关。Objective: To explore the inhibitory effect and mechanism of Shenshuai Granule (SSG) on transforming growth factor β1 (TGF β1) and tissue inhibitor of metalloproteinase 1 (TIMP 1) mRNA expression in remnant kidney. Methods: The model rats of 5/6 nephrectomized were prepared and treated separately with SSG, asiabell root, red sage and rhubarb by gastrogavage for 1 month. The levels of TGF β1 and TIMP 1 mRNA expression in the remnant kidney were determined using reverse transcription PCR. Results: TGF β1 and TIMP 1 mRNA showed over expression in the model group, but after 1 month treatment of SSG, the expressions declined significantly (P<0 01). The single drugs consisting of SSG, red sage and rhubarb showed inhibitory effect to certain degree, but asiabell root showed no effect. Conclusion: The effect of SSG in ameliorating renal fibrosis may be correlated with its inhibition on over expression of TGF β1and TIMP 1 mRNA in remnant kidney.
