保肾片对系膜细胞机械性伸展诱导的增殖因子和细胞外基质成分的影响

摘　　要：目的 :在肾小球硬化的进展中 ,肾小球内压升高是一种主要的启动因子 ,小球内压升高能增加系膜细胞的伸展强度。本研究探讨培养的大鼠系膜细胞中 ,机械性伸展对TGF - β和细胞外基质成分mRNA表达的影响 ,以及中药保肾片的抑制作用。方法 :用机械性伸展装置刺激培养的肾系膜细胞 ,并在培养液中加入喂服中药保肾片大鼠的血清 ,用Northernblot分析TGB - β1、3 及Ⅰ型、Ⅳ型胶原mRNA的表达。结果 :机械性伸展以时间依赖性方式刺激TGF - β1和TGF - β3 的mRNA表达 ,同时也发现机械性伸展能诱导系膜细胞外基质的主要成分Ⅰ型、Ⅳ型胶原及纤维结合素的mRNA表达亢进 ;中药保肾片对系膜细胞机械性伸展诱导的细胞增殖因子TGF - β1和细胞外基质主要成分Ⅰ型、Ⅳ型胶原的mRNA表达具有明显的抑制作用。结论 :机械性伸展刺激能够诱导产生TGF - β1、3 及Ⅰ型、Ⅳ型胶原的mRNA表达。中药保肾片延缓慢性肾衰竭的作用机理与抑制细胞增殖因子和细胞外基质成分mRNA的表达有关。Objective:To study the effect of mechanical stretch on the mRNA expression of transforming growth factor -β(TGF-β),and extracellular matrix(ECM)of cultured mesangial cells,and the suppressive effect of Chinese Herbal Baoshenpian tablet on them.Methods:The cultured kidney mesangial cells were stimulated by mechanical stretch device.Serum of the rats fed with Baoshenpian tablet was added to the culture medium. Northen blot was used to analyse the mRNA expression of TGF-β 1、3 ,and typeⅠ、Ⅳ of collagen.Results:The TGF-β 1,TGF-β 3 mRNA expression were up regulated by mechanical stretch in time-depending manner.Meanwhile hyperactive expression of Ⅰ,Ⅳ type of collagen and fibronectin were detected as well.Chinese Herbal Baoshenpian tablet exeres observable suppressive effect on the stretch-stimulated mRNA expression of TGF-β 1,and Ⅰ、Ⅳ type of collagen.Conclusion:The mechanical stretch can induce the mRNA expression of TGF-β 1、3 , and type Ⅰ,Ⅳ of collagen. The funetioning mechanism of Chinese Herbal Baoshenpian tablet in delaying the progress of Chronic Renal Failure is related to the restraint of the proliferative factors and ECM mRNA expressiom .
