摘 要:百慕大是一种难以培养的草坪草。本研究首次以百慕大成熟胚为外植体,通过胚性愈伤组织的发生进行植株再生。结果表明,百慕大成熟种子的出愈能力很强,且不同的基本培养基诱导效果相近,但初生愈伤组织呈无定型棉絮状,不能再生成株。采用提高培养基中蔗糖和琼脂浓度以及在培养基中附加适宜浓度ABA、CH和一定比例的生长素与细胞分裂素的方法,对初生愈伤组织实施继代改造,高频诱导出致密颗粒状的、具有高度再生能力的胚性愈伤组织,此类胚性愈伤组织在含有2.0mg/L6 BA、1.0mg/LKT和0.5mg/LNAA的分化培养基上再生频率较高。Bermudagrass was an important turfgrass and recalcitrant in tissue culture in generally. Embryogenic callus induction and plant regeneration were attempted for the first time from mature embryos of bermudagrass in this study . The results showed that high callus induction frequency was obtained from three different basic media, and the initial callus appeared spongelike, soft and irregular, from which no plants could be regenerated. After subculture and improvement of the initial callus by enhancement of sucrose and agar concentration, supplement of appropriate concentration of ABA and a suitable ratio of auxin/cytokinin in N6 medium, compact granular and high regeneration ability embryogenic callus were induced with high frequency. Green plantlets were substantially regenerated from the embryogenic callus on differentiation medium supplemented with 2.0 mg/L 6BA?1.0 mg/L KT and 0.5 mg/L NAA.
