摘  要:应用模糊数学方法。研究了不同浓度的2,4-D、KT、BA、ABA、NAA和IAA以及2,4-D+KT,2,4-D+BA,2,4-D+ABA+LH和NAA+IAA等多种附加物组合对水稻种胚离体培养的影响,目的在于对水稻种胚离体培养诱导培养基进行优化选择.结果表明,一种附加物的作用效果明显不如2种或3种附加物配合使用的效果好。以添加2,4-D 2.0mg/L+ABA 5.0mg/L+LH 300.0mg/L(U49)的改良MS培养基效果最好.研究还采用不同类型的水稻品种加以验证,表明大多数水稻品种的种胚离体培养于U_(49)组合的改良MS培养基上,可获得较高的愈伤组织诱导率,且有利于愈伤组织的再分化.In order to develop an efficient callus induction and regeneration system for rice breeding, mature embryo culture of rice was investigated to promote its efficiency by the use of supplements for callus induction medium combined with fuzzy mathematical analysis. Callus induction was carried out on modified MS medium (Murashige et al, 1962, Vit B_1 5.0 mg/ L, sugar 5.0%, agar 0.65%, pH 5.8) with different supplements or supplement combinations, after sterilization of seeds. Following supplements were studied, i.e. 2, 4-D (0.5, 1.0, 1.5, 2.0 mg/L), NAA (0.5, 1.0, 2.0, 4.0 mg/L), IAA (0.5, 1.0, 2.0, 4.0 mg/L), ABA (1.0, 2.5, 5.0, 10.0 mg/L), KT (0.5, 1.0, 1.5, 2.0 mg/L), BA (0.5, 1.0, 1.5, 2.0 mg/L), 2,4-D (1.0 or 2.0 mg/ L)+KT (1.0 or 2.0 mg/L), 2, 4-D (0.5 or 1.0 mg/ L)+BA (0.5 or 1.0 mg/ L),2, 4-D (0.5, 1.0, 1.5 or 2.0 mg/L)+ABA (1.0, 2.5, 5.0 or 10.0 mg/L)+LH (300.0 mg/L) and NAA (0.5 or 1.0 mg/ L)+IAA (0.5 or 1.0 mg/L). Mature embryos extracted from seeds of five rice cultivars (purelines) were in vitro cultured. After 30 days of culture, the calli were calaulated and then transferred into 1/2 MS medium supplemented with 2.0 mg/L BA, 0.25 mg/L IAA and 0.25 mg/L NAA. The results showed the effects of supplement combinations were better than supplements. The best one was modified MS medium with the supplements of 2, 4-D 2.0 mg/L+ABA 5.0 mg/L+LH 300.0 mg/L (U_(49). And further, 28 rice cultivars (purelines) were used to evaluate this result. The observation revealed that more calli was got on U_(49) medium in most cases. After transfer to regeneration medium, their green plantlet regeneration efficiency was higher as well.
