摘 要:黄色提取物经液相层析分离获得分子量为20~25kda的黄芪多糖,将其注射于14~16月龄的小鼠,可使其脾细胞在体外培养早期产生的白细胞介素2(IL-2)显著增加(P<0.05)。黄芪多糖的体内应用可使老年小鼠脾细胞培养中丝裂原激活的淋巴细胞对外源性IL-2的增殖反应显著增强(P<0.001),超过未给黄芪多糖的青年小鼠的反应水平。黄芪多糖通过促进脾细胞产生IL-2及增强脾细胞对IL-2的增殖反应改善老年小鼠的免疫功能。The extracts of Astragalus membranaceus were further seperated by liquid chromatography. The third fraction had been identified as a 20~25 kda polysaccharide (Astragalan). Groups of C57BL/6 mice were injected ip with Astragalan or normal saline (as control) for 5 consecutive days. The mixture of spleen cells prepared from each group of mice was cultured in the presence of Con A and TPA. In early culture (12 hours), the splenic lymphocytes of Astragalan-treated old mice, aged 14~16 months, produced higher amount of IL-2 than that of non-Astragalan-treated ones (P<0. 05). The level of IL- 2-stimulated proliferation of the spleen cells from Astragalan-treated ageing mice was significantly high- er than that from non-Astragalan-treated ageing mice (P<0. 001) or young mice (P<0. 01). These results suggest that administration of Astragalan is capable of improving the immunol function of ageing mice by enhancing the activities of their spleen cells in IL-2 production and IL-2-stimulated proliferation.
