摘　　要：目的观察重组水蛭素对Beagle犬凝血功能的影响。方法Beagle犬缓慢静脉注射不同剂量重组水蛭素 [6、18、5 4mg/(kg·d) ],连续 14d ,观察其凝血功能的变化。结果用药组犬皮下有血肿 ,活化部分凝血活酶时间和凝血酶时间明显延长 ,但凝血酶原时间无明显变化 ,高剂量组的血小板聚集率有升高趋势 ,光镜下观察脾脏等实质脏器有出血灶。结论重组水蛭素抑制内源性凝血系统 。PurposeThe effects of recombinat hirudin (rhirudin) on coagulant properties were observed in Beagle dogs.MethodsThe dogs were treated iv with rhirudin (6 ,18 and 54 mg/kg) daily for 2 weeks.ResultsThe administration of rhirudin resulted in dose dependent prolonging of the activated partial thromboplastin time and thrombin time. No significant effects of rhirudin on prothrombin time and maximum platelet aggregation were observed. Light microscopic analysis revealed minimal to moderate hemorrhage in some organs (spleen, et al ) and subcutaneous hemorrhage at the site of injection in the normal dogs treated with rhirudin.ConclusionThe results suggested that the inner coagulation system was inhibited by rhirudin, which resulted in dose dependent hemorrahage in multiple inner organs.
