摘 要:目的 研究N 硝基 L 精氨酸 (NO2 Arg)在吗啡、二氢埃托啡精神依赖形成中的作用。 方法 采用小鼠腹腔注射吗啡、二氢埃托啡的方法建立小鼠精神依赖的条件性位置偏爱模型 ;采用荧光内过滤法测定小鼠脑内NO的含量 ;小鼠脑内cAMP/cGMP含量采用放射免疫法测定。结果 小鼠吗啡、二氢埃托啡精神依赖形成过程中脑内NO ,cAMP ,cGMP的含量显著上升 ;腹腔注射NO2 Arg 5mg·kg- 1 可抑制吗啡、二氢埃托啡精神依赖升高小鼠脑内NO ,cAMP ,cGMP含量的作用。结论 NO在易化阿片类药物精神依赖形成中可能有一定作用 ;NO2 Arg抑制吗啡。OBJECTIVE: To investigate the role of N-nitro-L-arginine (NO2Arg) in the development of psychical dependence induced by morphine and dihydroetorphine. METHODS: The psychical dependence model was established by subcutaneous administration of morphine or dihydroetorphine into mice. The content of NO was determined by the fluorescence intensity based on the absorption of a reaction product of N-acetylcystaine and nitric oxide (NACNO) overlapping the absorption of quinine sulfate. The contents of cAMP/cGMP were measured by radioimmunoassay. RESULTS: The contents of NO, cAMP and cGMP in mice brains were increased significantly after consecutive injection of morphine and dihydroetorphine. Intra-peritoneal injection of 5 mg·kg-1 NO2Arg inhibited the development of psychical dependence induced by morphine and dihydroetorphine and the increases of NO, cAMP and cGMP content in brains. CONCLUSIONS: NO2Arg may suppress the development of psychical dependence induced by morphine and dihydroetorphine in mice by inhibiting the synthesis of nitric oxide and regulating the cGMP/cAMP content.
